Macroporous resin,silica gel column chromatography,C18column chromatography,sephadex LH-20 gel column chromatography and semi-preparative high performance liquid chromatography performed to separate and purify the chemical constituents of Hypericum lagarocladum N.Robson and Selaginella moellendorffii.Hieron.Spectroscopic methods such as MS and NMR combined with physicochemical properties were applied in identifying the structures of the isolated compounds.A total of 71 compounds were isolated and identified,including xanthanone,polycyclic polyprenylated acylphloroglucinols(PPAPs),steroid,flavone,biflvone,triterpenoid and fatty acids,among which 2 compounds were new compounds.The screening for the antidiabetic activity of some compounds from H.lagarocladum N.Robson ware performed.56 compounds were isolated from H.lagarocladum N.Robson,among them,17xanthones are identified as 1,7-dihydroxyxanthone(1),3,4,5-trihydroxyxanthone(2),2,7-dihydroxy-1-methoxyxanthone(3),1,3-dihydroxy-7-methoxyxanthone(4),1,5-dihydroxy-8-methoxyxanthone(5),3,4-dihydroxy-2-methoxyxanthone(6),3,4-dihydroxy-5-methoxyxanthone(7),2,3-dimethoxyxanthone(8),and2,3,4-trimethoxyxanthone(9),lagarxanthoneA(10),1.6-dihydroxy-5-methoxyxanthone(11),2,7-dihydroxyxanthone(12),1.7-dihydroxy-4-methoxyxanthone(13),1,3,7-trihydroxy-2-(3-methyl-2-butenyl)-xanthone(14),6-deoxyisojacareubin(15),jacareubin(16),garcibracteamone A(17),in which compound 10 was new compound and was identified as lagarxanthone A by 1D and 2D NMR and MS analysis;24polyprenylated acylphloroglucinol derivatives were identified as hyperlagarone A(18),hyperpatulone E(19),hyperbeanol G(20),uralione D(21),tomoeone F(22),pyramidatone A(23),tomoeone A(24),tomoeone B(25),hyperbeanol C(26),hyperbeanol A(27),hypercohone G(28),attenuatumione G(29),uralodin B(30),chipericumin C(31),chipericumin D(32),chipericumin E(33),hyperpatulol C(34),hyperpatulol D(35),hyperpatulol E(36),hyperielliptone B(37),hyperielliptone H(38),hyperbeanol B(39),hypascyrin D(40),hyperpatulone F(41),in which compound 18 is new compound and was identified as hyperlagarone A by 1D and 2D NMR and MS analysis;5 flavonoids and flavonoid glycosides were identified as quercitrin(42),apigenin-7-O-β-D-glucoside(43),apigenin-7-O-β-D-(6’’-O-acetyl)-glucoside(44),loteolin(45),quercetin(46);3triterpenoids were identified as betulinic acid(47),methyl betulate(48),betulonic acid(49);2 steroids were identified asβ-quebrahol(50),sitosterol-3-O-β-D-glicopiranosideo(51);3 organic acids were identified as ferulic acid(52),3,4-dihydroxy ethyl benzoate(53),benzoic acid(54),2 fatty acid ester(E,E)-9-oxooctadeca-10,12-dienoic acid(55)、phospholipase A2(56).We performed the antidiabetic on partial compounds.The result showed that compounds 17,31,32,34,38,40 exhibited significant effect on glucose uptake by 1.99,2.82,2.69,2.01,2.14 and 2.08,while other compounds exhibited weak effect on glucose uptake.15 compounds were isolated from S.moellendorffii.Hieron,in which 4biflavones were identified as ginkgetin(1),isoginkgetin(2),amentoflavone(3),(2R)-2,3-dihydroamentoflavone(4),2 flavonoids were identified as5-carboxymethyl-3′,4′,7-trihydroxyflavone(5),naringenin(6);one chalcone is identified as 3′,4′,7-trihydroxychalcone(15);five organic acids were identified as ethyl p-hydroxycinnamate(9),ethyl ferulate(10),vanillic acid(11),ethyl caffeate(12),cinnamic acid(13),p-coumaric acid ethyl ester(14);2 other compounds were identified as(3R)-thunberginol C(7),N-benzoyl-L-phenylalaninol(8).
