| Cholecystitis is a common gallbladder disease,and its incidence rate is only acute appendicitis.The main cause is secondary gallbladder inflammation caused by bile duct or bile obstruction.The common symptom is strong pain in the right upper abdomen.Cholecystitis is reported to cause gallbladder cancer,pancreatic cancer and colorectal cancer.At the same time,the prevalence of cholecystitis will also increase with age.Therefore,it is very necessary to seek effective means to treat cholecystitis.At present,the treatment methods of cholecystitis are surgical treatment and drug treatment,but the surgical treatment has some limitations,and the complications of some patients do not support surgical treatment.The drugs in clinical application are also relatively limited,so it is very necessary to develop new drugs to treat cholecystitis.Tibetan medicine has many years of clinical practice in the diagnosis and treatment of cholecystitis,with less adverse reactions and good curative effect on the whole.This paper discusses the pharmacodynamic effect and mechanism of Tibetan medicine Fu-fang-gan-qiang powder on cholecystitis from various angles.Fu-fang-gan-qiang powder is a Tibetan medicine compound developed by experts of Tibet Tibetan Hospital Based on many years of clinical experience in Tibetan areas.It has the functions of heat clearing,detoxification and promoting bile flow.Although Fu-fang-gan-qiang powder has been proved to be effective in the treatment of cholecystitis,its efficacy and mechanism have not been fully clarified.Therefore,this topic will explore the potential pharmacological effects of the drug from many angles and aspects.Using the research method of network pharmacology,the targets of chemical components in Fu-fang-gan-qiang powder and related targets of cholecystitis disease were intersected,and their network analysis was carried out.Then,the main chemical components and their contents in Fu-fang-gan-qiang powder were studied by HPLC analyzer.Finally,the efficacy and related molecular mechanism of Fu-fang-gan-qiang powder in the treatment of cholecystitis were explored through animal experiment in vivo and cell experiment in vitro.Through the network pharmacological analysis,it is preliminarily predict that the potential targets of Fu-fang-gan-qiang powder in the treatment of cholecystitis are IL-6 and IL-1βand TNF-αthe related signal pathways may be MAPK and PI3K-Akt signal pathways.Through the analysis of the chemical components of Fu-fang-gan-qiang powder,the main components and their contents are obtained.At the same time,these components are related to the treatment of cholecystitis.In the in vitro experiment of cholecystitis in guinea pigs,B-ultrasound was performed on guinea pigs in each group.It was observed that Fu-fang-gan-qiang powder could alleviate the thickening of gallbladder wall in guinea pigs with cholecystitis;The liver function indexes(alt,AST,TBIL,DBIL,ibil and TBA)in serum and the main indexes(TBIL,TC and Ca2+)in bile of guinea pigs in each group were detected.It was observed that Fu-fang-gan-qiang powder could down regulate various indexes to a certain extent;He staining of gallbladder and liver tissues of guinea pigs in each group and detection of the expression of relevant inflammatory factors in their tissues can reflect that Fu-fang-gan-qiang powder has a certain effect on the treatment of cholecystitis;Western blot showed that Fu-fang-gan-qiang powder could down regulate the levels of p-ERK 1/2,p-p38 and p-JNK in gallbladder tissue.In vitro cell experiment,it was found that Fu-fang-gan-qiang powder could significantly inhibit the expression of proinflammatory factors in cells and down regulate the levels of p-ERK 1/2,p-P38 and p-JNK in inflammatory cells.In conclusion,Fu-fang-gan-qiang powder has a good therapeutic effect on cholecystitis by inhibiting the activation of MAPK signal pathway,reducing the activity of inflammation related cells and the expression of inflammatory factors.Through the study of its efficacy and mechanism of action,it provides an experimental basis for the development of new drugs for the treatment of hepatobiliary diseases,and provides a reference for the later research of the drug. |
