Objectives:Under different experimental conditions,the behavior test,sleep observation and tongue image analysis of rhesus monkeys were carried out to explore whether the changes of the environment could affect the emotional changes of rhesus monkeys and the changes of tongue image caused by emotional changes,to provides theoretical basis for screening targets of tongue diagnosis of emotional diseases in TCM.Methods: Eight non-human primate rhesus monkeys(maraca mulatta)underwent four-wave behavioral tests and nocturnal exercise(sleep)under different experimental conditions.In the first wave,eight macaques were tested for their behavior and sleep within a social group.In the second wave of the experiment,eight macaques were subjected to the experimenter’s intrusion test and their behaviors were recorded,as well as their behavior and sleep status within the social group.In the third wave of the study,eight macaques were subjected to open field test,experimenter invasion test,and behavior recording,as well as the behavior and sleep status of the macaques within the social group.In the fourth wave,the behavior and sleep status of the eight macaques in the single cage were collected.The tongue images of eight macaques were collected and analyzed,and the behavior of the monkeys recorded in the video was analyzed.The behavior and sleep of the monkeys were quantified,and the state of the monkeys was judged by using statistical knowledge,to explore whether the changes of the environment could affect the emotional changes of the monkeys.Results:(1)In the first round of test,the monkey(No.18057)showed more stereotypic behaviors,and there was no significant difference in other behaviors among macaques.There was considerable variability in total sleep duration among the eight individual macaques.(2)In the second round of test,the monkey(No.18055)had the highest following percentage in the experimenter’s invasion test,and the monkey(No.18057)had the lowest following percentage.There was a difference between 18055 and 18057.18053 the monkey of social behaviors was relatively less,and the monkey of social avoidances was the most.18057 The monkey of stereotyped behaviors was the highest.18059 active social behavior for a long time;18053 longer durations of social avoidance;18057 spent the shortest time alone.18057 Stereotyped behavior was the longest.(3)Round 3: There was no difference among the eight monkeys in open field test.The following percentage was the lowest in the monkey(No.18331)and the highest in the monkey(No.18055).There were differences between 18055 and 18331.18059 the monkey of social behaviors was relatively high;18057 the monkey of solitary behaviors was relatively high;The monkey of active aggressions was relatively high.18053 The monkey of stereotyped behaviors was relatively high.18059 Active social behavior was relatively long;18057 Solitary behavior was relatively short;18057 The duration of stereotyped behavior is relatively long.18085 The duration of waking was relatively long,and overall there was little difference in sleep state among the eight macaques.(4)The tongue manifestation showed that all the eight monkeys were in pathological condition,with different syndrome types and severity.There were pathological factors such as heat,dampness and blood stasis in the body.(5)Single cage test: 18053 was the shortest.18055 had the shortest response time to the environment.The duration of light sleep was the longest in 18055 and the shortest in 18079.18085 deep sleep duration was the shortest;The total sleep duration of 18085 was the shortest.The duration of waking18085 was relatively long;The monkey of waking up was the least in 18085.The sleep latency of 18085 was the longest.(6)Macaque 18055 showed no significant difference between the second and third round of experimenter’s invasive behavior test,while all other monkeys showed significant difference.(7)There was no significant difference in the behavioral tests in macaque 18059,while all other macaques showed significant differences.(8)Compared with the four sleep tests of macaques,the sleep of eight macaques decreased in the third round of test.After the single cage test,the sleep of macaques 18053,18055,18057,18059,18083 and 18331 improved significantly,but could not recover as before.Macaque 18079,18085 sleep did not improve.(9)After the third round of test,the difference score showed that the overall state of the macaque decreased.After returning to the living single cage,the overall state of the macaque was significantly improved.Conclusions:(1)After experiencing the constant changes of the external environment,the monkeys showed abnormal behavior and pathological tongue manifestation.(2)Eight macaques returned to the original single cage,the macaques could not recover to the original state,indicating that the macaques suffered from continuous emotional stimulation.(3)Macaques adopt stereotypic behaviors to alleviate the effects of emotional stimuli.
