| Objects:By observing the efficacy of Uricqing granules in the treatment of patients with Gout syndrome of spleen deficiency and dampness,16 Sr DNA high-throughput gene sequencing technology was used to analyze the structure of intestinal flora(Lntestinal flore)in patients with Gout syndrome of spleen deficiency and dampness(IF)and the changes of intestinal flora structure before and after treatment of gout syndrome of spleen deficiency and dampness.The effect of Yuqing Granules on intestinal flora in patients with spleen deficiency and dampness obstruction of gout was preliminarily discussed,which provided scientific basis for clinical application of Yuqing granules and sought a new breakthrough in the treatment of gout with traditional Chinese medicine.Methods:Patients and healthy people meeting the inclusion criteria were divided into treatment group(32 cases)and control group(30 cases).The treatment group was given uricqing granules for 12 weeks.The changes of various indexes before and after treatment were observed and analyzed with statistics.At the same time,fecal samples before and after the treatment group and control group were collected by 16 s RNA high-throughput sequencing technology to establish a database,and the α diversity,βdiversity and intestinal flora composition structure were analyzed.On the one hand,the characteristics of the flora in the treatment group and the control group were compared,and on the other hand,the characteristics of the flora changes before and after the treatment group were compared.Results :1 Analysis of clinical efficacy results:After treatment with Uricqing granules,the decreased levels of UA,ESR and CRP indexes in patients with spleen deficiency and dampness syndrome of gout after treatment were statistically significant(P < 0.05),while TC,HDL-C,LDL-C and GLU were not statistically significant(P > 0.05),only TG was statistically significant P < 0.05).There was no significant difference in the physicochemical indexes of safety of WBC,RBC,Hb,PLTALT,AST,BUN and Cr between the two groups before and after treatment(P > 0.05).During the treatment,there were 2 patients with elevated ALT and AST indexes and 1 patient with elevated BUN indexes,and the adverse reaction rate was 9.37%.2 Analysis of intestinal flora results:The average total sequencing volume of all samples is 24729 tags,the average effective sequence volume is 24449 tags,the average sequence length is 1453 bp,the average total effective rate is 98.88%,the sequencing depth is reasonable,the sample size is sufficient.The number of OTUs,the total number of OTUs species,α diversity index,β diversity index and the difference analysis between the treatment group and the healthy control group were significantly different.There were significant differences in α diversity index,β diversity index and inter-group difference analysis before and after treatment,and there were differences in biodiversity and richness,as follows:(1)Compared with the healthy population,the patients with the syndrome of spleen deficiency and dampness of gout:(1)at the gate level:The average abundance of Firmicutes,Actinobacteriota and Proteobacteria in patients with spleen deficiency and dampness obstruction syndrome of gout was high.Proteobacteria and Firmicutes occupy the highest abundance of human intestinal flora(Lntestinalflore(IF)).Instead,the average richness of Bacteroidota and Desulfobacterota is low.Among all the flora with changed abundance,there were significant differences in Firmicutes,Proteobacteria and Bacteroidota except that Firmicutes,Proteobacteria and Bacteroidota had no significant differences(P < 0.05).(2)At the genus level: the average richness of Strptococcus,Lachnoclostridium and Blautia in gout patients with spleen deficiency and dampness was high.The average richness of Holdemanella was low.Among all the bacteria with altered abundance,there were significant differences in all bacteria groups except Faecalibacteria(P > 0.05).(2)Comparison before and after the treatment of spleen deficiency and dampness blocking syndrome of gout:(1)at the gate level: after treatment,the average abundance of Proteobacteria and Verrucomicrobia was high;Desulfobacteria,on the other hand,is low in average richness in Bacteroidota,Firmicutes,Fusobacteria,Patescibacteria and Desulfobacteria.In all the flora with changed abundance,except Proteobacteria and Firmicutes,there were significant differences in other flora(P < 0.05).(2)At the genus level,Meganmonas,Lachnoclostridium,Agathobacter and Akkermansia had high average richness after treatment;The average richness of Bacteroides,Dialister and Dorea was low.There were significant differences in all flora with changed abundance except Faecalibacteria,Strptococcus and Subdoligranulum(P < 0.05).Conclusion:1 Uric acid Qing granules can improve the clinical effect of gout syndrome of spleen deficiency and dampness obstruction.2 Uricuqing Granules have good safety in the treatment of gout patients with spleen deficiency and dampness obstruction,which can be further studied and promoted clinically.3 Compared with the healthy population,the patients with spleen deficiency and dampness syndrome of gout showed changes in intestinal flora diversity and decreased species richness.4 After taking Uricqing granules,intestinal flora types and richness of patients with spleen deficiency and dampness syndrome of gout can be adjusted. |
