Study On Modern Pharmaceutical Research Of HuoPu Xia Ling Decoction

Objective: To maintain the principle of following but not restricting to ancient exploration of the preparation process of Huopu Xia Ling granules,including extraction process as well as molding process study,establishing the quality standard of Huopu Xia Ling granules as well as the stability study,applied network pharmacology as well as the pharmacokinetic study on the basis of molecular docking.This study provides a foundation for the stability,reliability,and effectiveness of Huopu Xia Ling granules.Methods: The content and fingerprint similarity of amygdalin,honokiol,and honokiol as well as the extraction process were evaluated by five indexes baseline relatedness method combined with hierarchical analysis entropy weight method;Based on the idea that the quality is derived from the design concept,the Plackett Burman test design was used to screen the key process parameters of the molding process of the formulation,using the forming rate,moisture absorption rate and angle of repose as evaluation indicators;Box Behnken test was designed to optimize the molding process of the granulate by hierarchical analysis critical blending weighting method;Quality standards as well as stability studies of Huopu Xia Ling granules were carried out using thin layer chromatography as well as high performance liquid chromatography;Network pharmacology as well as molecular docking was applied to conduct the basic pharmacodynamic study.Results: the extraction process of Huopu Xia Ling granules was as prescribed amounts of medicinal materials,plus seven times the amount of water,soaked for 90 min,and decocted 1 time,each time for 1 h;The key process parameters of molding were screened out as ethanol concentration,ethanol dosage as well as drug / CO ratio;The molding process used dextrin as the excipient,the excipient ratio 1:1,ethanol concentration 80%,ethanol dosage 35%,drying at 55℃,whole pellets;Quality standards six medicinal herbs thin layer identification methods were established and the content of amygdalin,magnolol,as well as honokiol was determined;The long-term accelerated stability experiments in stability inspection items are well specified;Network pharmacology and molecular docking lay a certain foundation for further research on the mechanism of Huopu Xia Ling Decoction against liver injury.It guarantees that the preparation process of Huopu Xialing granules is stable and reliable and reproducible.
