Objective: To investigate whether autophagy and macrophage polarization occur in the livers of mice infected with Echinococcus granulosus and Echinococcus multilocularis,and to determine the effects of Echinococcus granulosus and Echinococcus multilocularis infection on autophagy-related proteins and macrophage polarization in the liver of mice.Methods: 1)to establish a mouse model of two types of Echinococcus infection through hepatic portal vein.2)blood samples were collected to detect the biochemical indexes of liver function.3)HE staining was used to observe the infiltration of inflammatory cells and the pathological changes of liver in the early stage of infection.4)Flow cytometry detection of Kupffer cell polarization.5)the expression of autophagy-related proteins LC3 and P62 was detected by immunohistochemistry and Western blotting.6)laser confocal microscopy was used to observe the number of autophagosomes in liver.Results: 1)the successful rate of two types of hydatid hepatic portal vein infection in mice was more than95%,and there was little effect on liver function related biochemical indexes in the early stage of hydatid echinococcosis(p > 0.05).2)the results of HE staining showed that there were differences in inflammatory cell infiltration and pathological phenotype between AE and CE mice at different time points of infection.Compared with CE,AE showed faster growth and more severe infection.3)the number of macrophages recruited from the liver of mice infected with Echinococcus multilocularis was different from that of mice infected with Echinococcus multilocularis(CE).Compared with mice infected with CE,the liver of mice infected with Echinococcus multilocularis multilocularis had a stronger ability to accumulate macrophages in the early stages of infection(p < 0.05).4)the expression of autophagy-related proteins LC3 and P62 in the liver of mice infected with Echinococcus multilocularis multilocularis was significantly higher than that of mice infected with CE(p< 0.05).5)the expression of LC3 and P62 in the liver of mice infected with Echinococcus multilocularis multilocularis multilocularis was significantly higher than that of mice infected with CE(p < 0.05),while the expression of P62 protein was not significantly different from that of mice infected with Echinococcus multilocularis multilocularis multilocularis multilocularis multilocularis multilocularis multilocularis(P > 0.05).Conclusion: 1)autophagy and macrophage polarization do exist in the early stage of Echinococcus granulosus infection,2)compared with CE,AE has stronger autophagic activity... |
