Objective: To study the effect of total ginsenoside(GS)on regulating lipid metabolism in mice fed with high-fat diet and its internal mechanism,so as to provide scientific basis for the development of new drugs with anti-obesity effect of total ginsenoside and the development and utilization of ginseng resources.Methods: High fat diet(HFD)mice were given GS(25,50,100 mg/kg/d)by gavage for 8 weeks.The weight,intake of water and random blood glucose,glucose tolerance and insulin tolerance were recorded.The changes of visceral adipose tissue(e WAT),subcutaneous adipose tissue(i WAT)and brown adipose tissue(BAT)were recorded at the end stage to determine the dose-effect relationship of GS.The pathological changes of e WAT and BAT in the 100 mg/kg/d dose group were observed by HE staining,the level of liver lipids was observed by oil red O staining,the level of serum and fecal lipids and the level of liver injury markers were detected by biochemical methods,the level of energy metabolism and solid gastric emptying were monitored,and the level of food intake related hormones was detected by ELISA kit.Serum non-target metabonomics was used to evaluate the variety and content of metabolites in mouse serum.Results: GS can significantly inhibit the weight growth rate of HFD mice in a dose dependent manner,improve the disorder of glucose and lipid metabolism in mice,and reduce the accumulation of peripheral and visceral fat in DIO mice.Further analysis revealed that GS may have an anti obesity effect by inhibiting mouse food intake,delaying gastric emptying,and promoting GLP-1 secretion.Finally,using metabolomics and metabolic phenotype analysis,it was found that the main mechanism of GS in anti obesity is its regulation of amino acid metabolism.Conclusion: Through technical means such as metabolomics,Micro-CT,and small animal constitution analysis,and using various molecular biological techniques,it was found that GS can improve lipid metabolism and fight obesity by regulating amino acid metabolism,inhibiting dietary intake,and promoting lipid metabolism in DIO mice.
