Expression And Significance Of MiRNA-574-5p,PIGF And SFlt-1 In Plasma Of Preeclampsia Patients

Objective:To explore the relationship between the plasma levels of micro RNA-574-5p(mi RNA-574-5p,mi R-574-5p),placental growth factor(PIGF),and its specific receptor soluble fms-like tyrosine kinase-1(soluble fms-like tyrosine kinase-1,s Flt-1)in patients with preeclampsia(PE)and their effects on PE development and clinical implications.Methods:From October 2021 to October 2022,60 patients with PE who had regular prenatal checkups and hospital deliveries at the People’s Hospital of Inner Mongolia Autonomous Region were collected as a case group,and were divided into PE group(n=30)and severe preeclampsia(s PE)group(n=30)according to the severity of the disease,and 30 healthy women who delivered in the same period at the hospital were selected as the control group.Thirty healthy women delivered in the same period in the hospital were selected as the control group.The expression level of mi R-574-5p in plasma was determined by quantitative real-time PCR(q PCR).Plasma concentrations of PIGF and s Flt-1 were determined by enzyme-linked immunosorbent assay(ELISA).Results:(1)The plasma mi R-574-5p expressions of pregnant women in the PE and s PE groups were 2.84±1.55 and 3.66±1.36,respectively,which were significantly higher than that in the control group of 1.03±0.50,and in the s PE group was again higher than that of the PE group(P<0.05).Plasma PIGF concentrations were36.25±5.31 pg/ml and 27.05±10.18 pg/ml,which were lower than 50.67±9.00 pg/ml in the control group,and lower in the s PE group than in the PE group(P<0.05).Their s Flt-1concentrations were 1923.95±213.69 pg/ml and 2331.62±189.59 pg/ml and their s Flt-1/PIGF ratio were 54.12±10.32,107.29±19.23,which were higher than those in the control one of1497.18±73.17 pg /ml and 30.44±5.54,all of the s PE groups were higher than the PE groups(P<0.05).(2)Plasma mi R-574-5p and PIGF levels of in PE were negatively correlated(r=-0.378,P<0.05).Plasma mi R-574-5p was positively correlated with s Flt-1 expression level(r=0.450)and s Flt-1/PIGF ratio(r=0.505)in PE(P<0.05).(3)APO occurred in 19 of 60 patients with PE,accounting for 31.66%.The APO ratios of mi R-574-5p and s Flt-1 high expression group were higher than the low expression group(P<0.05).the APO rates of the PIGF low-expressing group were higher than the high-expressing group(P<0.05).(4)Logistic regression analysis,the outcome showed that mi R-574-5p,PIGF,s Flt-1 level,and s Flt-1/PIGF ratio were independently affecting factors of PE-associated APO.(5)The area under the curve(AUC)of mi R-574-5p and PIGF,s Flt-1 level,and s Flt-1/PIGF ratio for predicting the occurrence of APO in PE were 0.823,0.935,0.865 and 0.946,and the AUC of s Flt-1/PIGF ratio combined with mi R-574-5p was 0.954,and the sensitivity and specificity were 82.90% and 98.00%,correspondingly.Conclusion:Plasma mi R-574-5p expression was elevated and PIGF expression was decreased in pregnant women with PE,and their s Flt-l express and s Flt-1/PIGF ratio were increased,and the degree of change in each index was associated with the seriousness of PE,may be implicated in the developing PE.This study also concluded that the combination of s Flt-1/PIGF ratio and mi R-574-5p had a certain contribution in judging the severity of the disease and as an auxiliary predictor of APO This step of judgment and prediction is crucial for clinical pregnancy monitoring and pregnancy termination decision.clinical pregnancy monitoring and pregnancy termination decision.
