| Objective:Through data mining technology,the literature of modern doctors using traditional Chinese medicine to treat visual fatigue was collated and analyzed,and the regularity of visual fatigue medication and core prescriptions were summarized and summarized.At the same time,network pharmacology is used to explore the potential mechanism of core prescription in the treatment of visual fatigue,providing some reference value for the rational use of drugs and new drug research and development in the clinical treatment of visual fatigue.Methods:The literatures of traditional Chinese medicine treatment of visual fatigue were searched in CNKI,Wanfang,VIP,Chinese biomedical literature database and Pub Med database,and the prescriptions were screened according to the inclusion and exclusion criteria.Collect the information of literature and prescription,establish the database of visual fatigue,and import the software of ancient and modern medical record cloud platform.Through association rule analysis,cluster analysis,complex network pharmacology analysis and other methods to explore the law of traditional Chinese medicine treatment of visual fatigue medication,to obtain the core prescription of visual fatigue.The core formula was searched through TCMSP database to obtain the main chemical components and related targets of action,and the obtained targets were standardized in Uniprot database,and the network diagram of herb-active component-target relationship was drawn through Cytoscape software to screen the main active components.Targets associated with visual fatigue were searched and screened in the Gencards and OMIM databases,and duplicates were removed after merging.The common target genes of visual fatigue and core were obtained by taking the intersection using Vinny software.String database was used to construct PPI networks and screen key target genes.GO enrichment analysis and KEGG enrichment analysis of the resulting targets were performed using the Bioconductor platform.Molecular docking of key target genes and major active components was performed using Auto Dock Vina software.Results:The data mining results of traditional Chinese medicine treatment of visual fatigue showed that 86 literatures were included,111 prescriptions,involving 162 herbs,of which 17 herbs were used with frequency≥20,and the top 10 herbs were Angelica sinensis,Radix Paeoniae Alba,Fructus Lycii,Radix Bupleuri,Radix Glycyrrhizae,Radix Rehmanniae Rehmanniae Rehmanniae,Rhizoma Chuanxiong,Poria cocos,Radix Codonopsis pilosulae The medicinal properties of traditional Chinese medicines for visual fatigue are mainly warm,flat and slightly cold;the five flavors are mainly sweet,pungent,bitter and sour;the meridians are mainly liver meridian,heart meridian and spleen meridian;and the most frequently used efficacy is clearing away heat and toxic substances.Association rules showed that Angelica sinensis–Paeonia lactiflora,Glycyrrhiza uralensis–Paeonia lactiflora had the highest co-occurrence.Cluster analysis showed that three drug groups were obtained,the first group: Bupleurum chinense,Glycyrrhiza uralensis,Kudzu root,Atractylodes macrocephala,Codonopsis pilosula,Astragalus membranaceus,and Cimicifuga racemosa;the second group: Lycium barbarum,Chrysanthemum morifolium,Rehmannia glutinosa,Poria cocos,Yam,and Cortex Moutan;and the third group:Angelica sinensis Complex network analysis showed that seven herbs,Angelica sinensis,Bupleurum chinense,Rehmannia glutinosa,Chuanxiong,Paeonia lactiflora,Lycium barbarum,and Glycyrrhiza uralensis,were finally obtained to form the core formula for the treatment of visual fatigue in traditional Chinese medicine.Pharmacological analysis of the core formula network showed that the main active components of the visual fatigue core formula included quercetin,kaempferol,β-sitosterol,stigmasterol,and isorhamnetin.The PPI network diagram showed that the key target genes mainly included AKT1,IL-6,TNF,JUN,and RELA.Molecular docking of the main active components and key target genes revealed that the binding energy of the main active components to the target protein was less than-5 kj/mol,suggesting that the two could stably bind.Conclusion:The data mining results reflect the clinical medication regularity of modern Chinese medicine in the treatment of visual fatigue and the basic pathogenesis understanding of visual fatigue based on virtual scale and mixed deficiency and deficiency,and summarize the core formula of asthenopia composed of seven herbs:Angelica sinensis,Bupleurum chinense,Rehmannia glutinosa,Chuanxiong,Paeonia lactiflora,Lycium barbarum,and Glycyrrhiza uralensis.Through network pharmacology analysis of the core formula,it is concluded that the core formula may regulate asthenopia by regulating inflammatory response,regulating apoptosis,improving oxidative stress,regulating retinopathy and other effects of intervention. |
