| Objective:To investigate the expression of CRABP1,ERα,ERβ and PR in PTC,and to explore the correlation between the expression of CRABP1,ERα,ERβ and PR and the clinicopathological characteristics,so as to provide a new reference for exploring the targeted therapy and prevention of PTC.Methods:1.Bioinformatics tools such as R software,Web Gestalt,annotation,Cluster Profiler,STRING and Cytoscape were used to identify the key genes and their correlations in PTC.2.The expression of CRABP1,ERα,ERβ and PR molecules in 101paraffin-embedded PTC tissues and corresponding adjacent non-cancerous tissues was detected by immunohistochemistry.The correlation between CRABP1,ERα,ERβ and PR molecules and the pathological characteristics of PTC and the correlation between the expression of each molecule were analyzed.3.Ten cases of fresh PTC tissues and corresponding adjacent tissues were collected.The mRNA and protein expression levels of each molecule were detected by RT-PCR and Western blot.Results:1.There are 308 differential Expression genes(DEGs)in the five data sets of Gene Expression Omnibus(GEO),CRABP1 is a key gene in PTC screened by PPI network.2.The expression of CRABP1 was negatively correlated with ERα and PR,and positively correlated with ERβ in PTC.3.The expression of CRABP1 in PTC was significantly lower than that in paracancer tissues(P <0.05),and was closely correlated with lymph node metastasis(P<0.05).The expression of ERβ in PTC was significantly lower than that in paracancer tissue(P < 0.05),and was closely related to lymph node metastasis and invasion of surrounding tissue(P<0.05).The expression of ERα and PR in PTC was significantly higher than that in paracancer tissue(P <0.05).It was closely related to lymph node metastasis and surrounding tissue invasion(P<0.05).The expression of CRABP1 was negatively correlated with ERα,while the expression of other proteins was not correlated.4.The results of RT-PCR and WB showed that the mRNA and protein expression levels of CRABP1 and ERβ in PTC were lower than those in adjacent tissues,while the mRNA and protein expression levels of ERα and PR were higher than those in adjacent tissues.Conclusion:The expressions of CRABP1 and ERβ are down-regulated,while ERα and PR are up-regulated in PTC,and the expression of CRABP1 is negatively correlated with the expression of ERα,which may provide a new research direction for the treatment of PTC. |
