| Objective:The first chapter aims to establish a fast,comprehensive and efficient capillary electrophoresis method for simultaneous separation and determination of Loureirin A,B,C and D with methanol and ethylene glycol as electrolyte additives.This method will provide a guidance for the quality control of Resina Draconis and its preparations.The second chapter aims to determine the p_values of four Loureirins by capillary zone electrophoresis.It will provide a basis for the study of the related reactions and pharmacokinetic characteristics of the analytes.Methods:1.The experiments were performed by using a BECKMAN P/ACE MDQ system equipped with a diode array detector.And an uncoated fused-silica capillary column(40.2cm×75μm id)with an effective length of 30 cm was used.The effects of several important experimental parameters were investigated,including the detection wavelength,the concentration of buffer,the p H of background electrolyte,the type and content of additives,separation voltage and temperature.The optimum separation and determination conditions were as follows:60.0 mmol/L sodium tetraborate(p H=9.60)as buffer solution,5.0%(v/v)methanol and 3.0%(v/v)ethylene glycol as additives,wavelength 329 nm,the injection pressure of 0.3 psi,the injection time of 3 s,separation voltage 17.5 k V,temperature 25℃.2.The experiments were carried out using a BECKMAN P/ACE MDQ system equipped with a diode array detector.And an uncoated fused-silica capillary column(40.2cm×75μm id)with an effective length of 30 cm was used.The p_of four Loureirins were determined by Origin nonlinear fitting method based on the effective mobility at different p H values under constant ionic strength.Results:1.Under the optimal electrophoretic conditions,Loureirins A,B,C and D could reach completely separation within 10 min.The linear relationships were good,and the correlation coefficients was in the range of 0.9937~0.9994.The peak area precision was0.71%~1.6%(intraday)and 1.3%~1.9%(interday),and the migration time precision was0.31%~0.75%(intraday)and 0.52%~1.5%(interday),respectively.The average recoveries of real samples were in the range of 98.4%~101.8%.2.The measured p_values for the four Loureirins were Loureirin A(8.01),Loureirins B(8.04),Loureirins C(8.24),Loureirins D(8.35),respectively.Conclusion:In this study,a simple,fast and efficient capillary electrophoresis method with organic solvent as additive was established for the separation and determination of four dihydrochalcones in Resina Draconis.Which was a promising alternative guidance for the quality control and development and application of the herbs and preparations of Resina Draconis.And a capillary zone electrophoresis method was established to successful determine the p_values of these four Loureirins in Resina Draconis.Which provided a reference for the determination of p_values of dihydrochalcones by capillary electrophoresis. |
