| Hyperlipidemia poses a serious risk to human health.Its incidence rate and mortality are increasing worldwide.In China,the overall prevalence of hyperlipidemia is as high as 35.6% among people aged 18 and over.Therefore,the treatment of hyperlipidemia is particularly important.Sanghuangporus sanghuang is a valuable edicine and food homology macrofungus,belonging to the genus Sanghuangporus of the family Hymenochaetaceae.Sanghuangporus sanghuang can help to decrease elevated blood lipids caused by lipid deposition,resulting in a lipid-lowering effect.Therefore,researching the lipid-lowering effect of Sanghuangporus sanghuang is of significant research value and importance.The purpose of this study was to investigate the effect of different extracts of Sanghuangporus sanghuang on lipid-lowering effect,using the value of lipid-lowering activity in vitro as an indicator.First,Ethanol reflux extraction was performed on the dried fruiting bodies of Sanghuangporus sanghuang to obtain the ethanol extract of Sanghuangporus sanghuang.Then,the ethanol extract of Sanghuangporus sanghuang is extracted with ethyl acetate and n-butanol to obtain the ethyl acetate extract and nbutanol extract of Sanghuangporus sanghuang,respectively.To investigate the lipidlowering effect of active ingredients in Sanghuangporus sanghuang,Hep G2 cells induced by oleic acid in vitro was used as the high-fat cell model.The TG and TC contents in the Hep G2 high-fat model cells were measured,and the lipid content determined by oil red staining assay.The morphological changes of the Hep G2 highfat model cells were measured by SEM,and the changes of cell mechanical properties were detected by AFM.These were used to investigate the active components of the lipid-lowering effect of Sanghuangporus sanghuang.The experimental results demonstrated that the ethyl acetate extract of Sanghuangporus sanghuang had a superior lipid-lowering effect compared to the n-butanol extract.Using lipid-lowering activity as the index,the ethyl acetate extract was fractionated into five fractions(H1-H5).Then the main components in H1-H5 were analyzed by LC-MS,and six components were analyzed.These included one kind of coumarin(H1-1: 7-methoxycoumarin),one kind of benzoic acid(H5-2: O-Toluic acid),two kinds of polyphenols(H2-1: Caffeic acid,H5-1: Protocatechuic acid),and two kinds of flavonoids(H3-1: 5-hydroxy-7-methoxy-flavone,H4-1: Naringenin).In the high-fat model cells induced by oleic acid,it was found that the lipid deposition,TG,TC content,and other indicators of the Hep G2 cells significantly increased.The Hep G2 high-fat model cells were bulging,and their height,adhesion force and Young’s modulus were significantly increased.However,after the treatment of fractions H1-H5,the lipid deposition and the content of TG and TC in the Hep G2 high-fat model cells were reduced,corrected the bulging shape of the cells and reduced the mechanical properties of in the Hep G2 high-fat model cells.Notably,the lipid-lowering effect of fraction H4(Naringenin)was most obvious.In summary,in this paper,it explored the active components responsible for the lipid-lowering effect of Sanghuangporus sanghuang,and found that Naringenin from the ethyl acetate extract had a more significant lipid-lowering effect in vitro.These findings enriched the study of active ingredients of Sanghuangporus sanghuang and provide valuable information for further research and application of its lipid-lowering activity.The experimental data obtained also provides a basis for the treatment of hyperlipidemia. |
