| Objective:To explore the effect of Yujing Jiangtang Recipe on glucose and lipid metabolism and oxidative stress in type 2 diabetes mellitus(T2DM)rats,and to provide some experimental basis for the rational development and clinical use of the drug.Methods:60 Sprague-Dawley(SD)rats of SPF grade were randomly selected as normal control group and fed with normal maintenance feed.The other 50 rats were fed with high-fat diet for 4 weeks,and then combined with intraperitoneal injection of 40 mg/kg Streptozotocin(STZ)to build T2DM rat model-Forty five T2DM rats were randomly divided into a diabetes model group,a metformin group(0.2 g/kg),a low dose group(1 g/kg),a medium dose group(2 g/kg)and a high dose group(4 g/kg)of Yujing Jiangtang Recipe,with 9 rats in each group.The treatment group was gavaged with corresponding dose,the normal control group and the diabetes model group were gavaged with 10 ml/kg normal saline according to weight,and each group was gavaged once a day for 6 consecutive weeks.During the experiment,observe and record the weight changes of rats in each group,and measure fasting blood glucose(FBG);After continuous gavage for 6 weeks,the abdominal aorta blood of rats was taken,the fasting insulin(FINS)level and the percentage content of HbAlc were measured,HOMA-IR and ISI were calculated;Serum TG,TC,HDL-C and LDL-C were measured;The serum levels of MDA,SOD,GSH,NO,GSH-Px and CAT were measured by ELISA;HE staining was used to observe the pathological changes of rat pancreas.Results:1.Effect of Yujing Jiangtang Recipe on the body weight of T2DM rats:After 6 weeks of administration,compared with the diabetes model group,the body weight of the high dose group of Yujing Jiangtang Recipe increased significantly(P<0.01).2.Effect of Yujing Jiangtang Recipe on blood glucose,FINS,HOMA-IR,ISI and HbAlc in T2DM rats:After 6 weeks of administration,compared with the diabetes model group,the FBG in each dose group of Yujing Jiangtang Recipe significantly decreased(P<0.01),and it can reduce FINS,HOMA-IR and HbAlc in rats,and increase the ISI content(P<0.05 or P<0.01).3.Effect of Yujing Jiangtang Recipe on blood lipid of T2DM rats:After 6 weeks of administration,compared with the diabetes model group,the levels of serum TG,TC,LDL-C in each dose group of Yujing Jiangtang Recipe decreased(P<0.05 or P<0.01),and the level of HDL-C increased(P<0.01).4.Effect of Yujing Jiangtang Recipe on the level of oxidative stress in T2DM rats:After 6 weeks of administration,compared with the diabetes model group,the content of serum MDA in each dose group of Yujing Jiangtang Recipe significantly decreased(P<0.05 or P<0.01),and the levels of serum SOD and GSH significantly increased(P<0.01);The levels of NO,GSH-Px and CAT in the middle and high dose groups of Yujing Jiangtang Formula were significantly increased(P<0.01).5.Effect of Yujing Jiangtang Recipe on pancreatic histopathology of T2DM rats:After 6 weeks of administration,compared with the diabetes model group,the exocrine acini in the high dose group of Yujing Jiangtang Recipe were more complete and regular in structure,the islet cells were more regular in morphology,and the number of cells in the islet was more,and the arrangement was even.Conclusion:Yujing Jiangtang Recipe has hypoglycemic effect on type 2 diabetes rats,and its mechanism may be related to regulating blood lipid metabolism,improving insulin resistance,antioxidation and repairing pancreatic tissue. |
