Objective The purpose of this study is to explore the pathogenesis of COPD-PAH and provide ideas for clinical early diagnosis and treatment by sequencing the circ RNA of COPD,COPD-PH and healthy control group and analyzing the differential expression according to the expression results.Methods Six COPD-PH patients who were treated in the respiratory department of the Ningxia People’s Hospital from December 2021 to July 2022 were included as the disease group,six COPD patients were included as the disease control group,and six healthy volunteers who were examined in our hospital at the same time were included as the health control group.The circ RNA in peripheral blood samples were sequenced by high-throughput sequencing technology,and the correlation of biological replicates was assessed after sequencing.For the experiments with biological duplication,DESeq2 was used for differential expression analysis,and the analysis results were analyzed by hierarchical cluster analysis.Finally,functional annotation and enrichment analysis were carried out for circ RNA differential source genes,including GO classification,Go Enrichment Hierarchy analysis,KEGG annotation,KEGG pathway enrichment analysis,the role of circ RNA in the development of COPD-PH and its value as a marker of early onset has been investigated.Results A total of 26401 circ RNA were screened by high-throughput sequencing.FC≥ 2 and P < 0.05 were used as screening criteria to detect 251 differentially expressed circ RNA in peripheral blood of patients with COPD-PH,including 164 up-regulated circ RNA and 87 down-regulated Circrnas.GO analysis and KEGG analysis of differentially expressed circ RNAs revealed the presence of MAPK pathway in the first 5 significantly enriched results,suggesting that the mechanism of action of circ RNAs in the development of COPD-PH may be related to MAPK signaling pathway.Conclusion A large number of circ RNAs are differentially expressed in peripheral blood of patients with COPD-PH,suggesting that circ RNA may be involved in the pathogenesis of COPD-PH.Functional enrichment of circ RNAs differentially expressed in peripheral blood of patients with COPD-PH indicated that circ RNA might play a role through MAPK pathway、apoptosis and other ways,which has the value of serving as early markers of COPD-PH...
