Corydalis hendersonii Hemsl.(CH),belonging to the family Papaveraceae,is mainly distributed in the western and middle Tibet at the altitude of 4200-5200 m.Its whole plants,a traditional Tibetan folk medicine,named "Ai-zi-jin" in Chinese have the functions on the treatment of Mubu related diseases,clearing heat,relieving pains,polycythemia vera and diarrhea in clinic which indicates that it might be related to the antinociceptivity,antiinflammatory and antiplatelet aggregation.Previous LC-MS analysis of the EtOH extract of CH revealed the main chemical constituents were isoquinoline alkaloids which exhibit a variety of pharmacological activities.Our analgesic evaluation of the ethanol extracts of CH in three animal models by hot plate,formalin and writhing showed that it had significant analgesic activity.Considering the clearly analgesic activity of Corydalis yanhusuo W.T,we hypothesize that the antinociceptivity of CH may be related to its alkaloid composition.In order to investigate the pharmocological consitituents of CH,a phytochemical study on the total alkaloids and BuOH parts were performed.Generally,this thesis is composed of the following sections:The review section summarized the alkaloids isolated from the genus Corydalis and a comprehensive summary on pharmacological progress,hoping to provide some guides in the research project.The phytochemistry section,as the major content of this thesis,presents a chemical investigation on the total alkaloids parts and BuOH parts of the EtOH extract of CH,respectively.On the basis of repeated silica gel,Sephadex LH-20,ODS,preparative HPLC methods and extensive spectroscopic data analysis including 1D,2D NMR,MS,and comparison of their NMR data with those reported,a total of 35 compounds were isolated and 32 of them were structural idenficatied.Among which,novel compound(1a and 1b),six new alkaloids including two benzylisoquinolines(2 and 7),four spirobenzylquinoline(3-6)were isolated.The next section is in vitro assays,which includes the anti-inflammatory activities based on the H9c2 cells,neuroprotective effect of rat adrenal phechromocytoma cell line(PC 12)induced by L-glutamic acid,antiplatelet aggregation induced by THR,AA and ADP.In brief,the present study covers a phytochemical research in addition to a preliminary in vivo and in vitro evaluation on CH.The results reveal that alkaloids are the main consituents which exhibite a varity of significant bioactivities.A total of 35 compounds including 8 new ones were isolated and structural identified.This thesis provides some phytochemical and pharmacological data for illustrating effective ingredients of CH and the genus Corydalis.
