Objective:To observe the proliferation of chondrocytes and the changes in the concentrations of IL-1 and MMP-3 in the supernatant of the rabbit’s knee osteoarthritis chondrocytes treated with serum containing Zhitong Jiangu Decoction,and to explore the effects of zhitong jiangu decoction on the proliferation of chondrocytes and the expression of IL-1 and MMP-3 in chondrocytes.To provide an experimental basis for the study of the mechanism of Zhitong Jiangu Decoction on KOA.Methods:After the rabbit KOA model was prepared successfully,chondrocytes of the rabbit knee joint were isolated and cultured by the enzyme digestion method,and the third generation of chondrocytes were subjected to grouping drug intervention.The experiment was divided into 7 groups.The blank control group did not receive serum,and the normal serum group and the Zhitong Jiangu Decoction-containing serum group were set at concentrations of 5%,10%,and 20% volume fraction,respectively.After serum intervention for 24 h,48h and 72 h in each group,the proliferation was detected by cck-8 method,and the concentrations of IL-1 and MMP-3in the supernatant of KOA chondrocytes were detected by ELISA method.Results:After 24 h,48h and 72 h of serum intervention,compared with the blank group at the same time point,the proliferation of chondrocytes was faster in the normal serum group and the drug-containing serum group(P<0.05).Compared the same volume fraction that between the normal serum group and the drug-containing serum group,the proliferation of drug-containing serum group was faster(P<0.05),and the effect of 20%drug-containing serum was the most significant(P<0.05),and the proliferation of chondrocytes was the most after 72 h of serum intervention(P<0.05).Compared with the blank group at the same time point,the concentration of IL-1 in the normal serum group and the drug-containing serum group decreased(P<0.05).The concentration of IL-1 in the drugcontaining serum group was lower than that in the normal serum group with the same volume fraction as that in the drug-containing serum group(P<0.05),and the effect of 20% drug-containing serum was the most significant(P<0.05).At 48 h to 72 h after the serum intervention,the drugcontaining serum still had the effect of reducing IL-1(P<0.05).Compared with the blank group at the same time point,the concentration of MMP-3decreased in both the normal serum group and the drug-containing serum group(P<0.05).The concentration of MMP-3 was lower in the drugcontaining serum group than in the normal serum group with the same volume fraction(P<0.05).In the normal serum group and the drugcontaining serum group,the concentration of MMP-3 in the 10% and 20%volume fraction serum group was lower than that in the 5% serum group(P<0.05).At 48 h to 72 h after the serum intervention,the serum still had the effect of reducing MMP-3(P<0.05).Conclusion:Zhitong Jiangu Decoction can delay the degeneration of knee cartilage by promoting the proliferation of the knee osteoarthritis chondrocytes and down-regulating the expression of IL-1 and MMP-3.
