Objective:Based on the Xuanfu theory and the target of inhibiting angiogenesis,observe the effect of Yinxieping Pills of "Cooling the blood and detoxification and opening Xuanfu" on the skin lesions of psoriasis-like mice induced by imiquimod(IMQ)cream Of VEGF and its receptor FLT-1and KDR expression levels.Methods:48 female BALB/c mice were randomly divided into 6 groups:blank control group,model control group,Avi A capsule group and Yinxieping pill low,medium and high dose groups,each with 8 mice.After the mice were anesthetized,the back hair was removed to form a 2cm*3cm skin exposure area.Except for the blank control group,which was externally coated with white petrolatum,the other 5 groups were externally coated with 5% imiquimod cream to induce psoriasis models,Once a day,the dose is 62.5mg,for 8 consecutive days.Gavage intervention was performed while administering topical drugs to mice.The blank control group and model control group were given 0.9% normal saline by gavage,and the Acitretin capsule group was given Acitretin capsule solution.Yinxieping pills were low,medium,and The high-dose group was given low,medium,and high-concentration Yinxieping pill solution by gavage,once a day,with a dose of 0.4ml for 8 consecutive days.24 hours after the last administration,the morphological changes of the skin lesions of the mice were observed and photographed by naked eyes,and scored by the area and severity index(PASI)of the skin;the skin lesions on the back of the mice were taken for pathological section analysis,and the immunohistochemical method was used to detect each The expression levels of VEGF and its receptor FLT-1 and KDR in the skin lesions of mice in the group.Results:(1)Compared with the blank control group,the PASI score of the skin lesions of the model control group increased significantly,and the difference was statistically significant(P<0.05).Compared with the model control group,the PASI scores of the skin lesions in the Acitretin capsule group and the Yinxieping pill low,medium and high dose groups were significantly reduced,and the difference was statistically significant(P<0.05).(2)The pathological changes of the skin lesions of mice in the low,medium and high doses of Yinxieping Pills were lighter than those of the model control group,and the degree of pathological changes of the skin lesions gradually decreased as the concentration of Yinxieping Pill solution increased;(3)VEGF and its receptors FLT-1,KDR were only slightly expressed in the skin lesions of the blank control group;compared with the blank control group,VEGF and its receptors FLT-1,KDR were expressed in the skin lesions of the model control group.The expression of KDR increased significantly,and the difference was statistically significant(P<0.05);Compared with the model control group,VEGF and its receptors were found in the skin lesions of the Acitretin capsule group and the Yinxieping pill low,medium and high dose groups.The expression of FLT-1 and KDR decreased significantly,and with the increase of the concentration of Yinxieping Pill solution,their expression decreased further,and the difference was statistically significant(P<0.05).Conclusion:(1)The Yinxieping Pills with cooling blood detoxification and Xuanfu legislation can significantly reduce the PASI score of psoriasis-like mouse model skin lesions,improve the symptoms of psoriasis-like skin lesions,and have a therapeutic effect on psoriasis.(2)Compared with the normal control group,the immunohistochemical expression levels of VEGF and its receptor FLT-1 and KDR in the skin lesions of psoriasis-like mice in the model group were significantly increased,indicating that the occurrence of psoriasis may be related to VEGF and The expression of its receptor FLT-1 and KDR are related.(3)Yinxieping pill can reduce the expression levels of VEGF and its receptor FLT-1 and KDR in the skin lesions of psoriasis-like mice.This may be one of the mechanisms of Yinxieping pill in the treatment of psoriasis by inhibiting angiogenesis.
