The Experimental Studies On Efficacy And Mechanism Of Yinxieping Granules For Psoriansis

ObjectiveTo investigate and evaluate the Efficacy, Mechanism and safety of Yinxieping Granule so as to bear out the effect of Yinxieping Granule on psoriasis and provide experimental basis for its clinical practice by observations of effect of YinXieping Granule on psoriasis in guinea pigs, and effects of YinXieping Granule on mitosis of vaginal epithelial cell in mice and on the formation of mouse-tail scale epidermis granular layer, acute toxicity test.Methods1.70 male guinea pigs were randomly divided into 7 groups. Guinea pigs in all groups except those in the blank group were smeared with 5% propranolol emulsion to develop psoriasis model. Yinxieling Granule, Methotrexate, and high, middle, low dose of Yinxieping Granule were administered into the guinea pigs in the different groups. The effects of the drugs on psoriasis, such as histopathological changes in skin lesions, hemorrheological changes, related cytokines TNF-a,ICAM-1 expression, were observed and detected respectively.2.50 ICR mice of half gender were randomly divided into groups. Yinxieling Granule, Methotrexate, and high, middle, low dose of Yinxieping Granule were orally administered into the mice in the different groups for 14 consecutive days. Then, the mice were sacrificed, and a piece of the tail skin was harvested and sliced. Count of the scales with granular layer was carried out under microscope.3.60 female mice were randomly divided into groups. Intraperitoneal injection of diethylstilbestrol was performed for 3 consecutive days in the mice in all groups except those in the blank group to induce the vaginal epithelium to be in estrus. Then, methotrexate, and high, middle, low dose of Yinxieping Granule were orally administered into the mice in the different groups, and an equal volume of distilled water was orally administered into the mice of the black control group and model group. Colchicine was orally administered into the mice 1 hour after the last administration to induce vaginal epithelial mitosis to be in the mid-term. After 8 hours vaginal tissue was taken, and count of mitosis was carried out under microscope.4.40 ICR mice of half gender were randomly divided into the treatment group and the blank control group. Yinxieling Granule (2.6g crude drug/ml) was administered into the mice of the treatment group at a dose of 0.4ml/10g, two times within 24 hours, and an equal volume of distilled water was administered into the mice of the black control group. The mice mortality and the general situation were observed and recorded for 14 consecutive days, and the mouse was weighed every day during observation.Results1. The results of effects of Yinxieping granule on guinea pig model of psoriasis showed the general shape of the guinea pigs in treatment group and their binaural morphology were remarkably improved as compared to those of the control group. Cytokine TNF-a and ICAM-1 levels in the treatment group were significantly lower than those in the model group, and the difference was significant.2. The mice tail scales with granular layer in Yinxieping granule group were more than those in the control group, and the difference was significant. Mitotic index of vaginal epithelial cells in mice of Yinxieping granule group was significantly lower than that in mice of the control group, and the difference was significant.3. During observing for 14 days, there was no death in all the mice, and the differences in body weight and general condition between the two groups were not significant. Conclusion:Yinxieping granule has definitely therapeutic effect on psoriasis by imploving the general state, histopathological, reducing the cytokine level and promoting formation of granular layer in mouse tail scale epidermis and inhibiting the mitosis of vaginal epithelial cells to improve the hyperkeratosis.Yinxieping granule has no acute toxicity in mice, which can be referenced for long-term toxicity for experimental study and clinical application.
