| Objective:(1)To collect literatures on the treatment and classification of lung cancer syndrome-types in recent 14 years and summarize the law of lung cancer syndrome-types;(2)To explore the effect of Jiaweifeifu decoction on tumor growth of lung adenocarcinoma bearing mice;(3)To explore the effects of Jiaweifeifu decoction on autophagy and AKT-m TOR signaling pathway of lung adenocarcinoma mice.Methods:(1)Relevant literatures on TCM syndrome differentiation and treatment of lung cancer published in domestic journals from January 1,2008 to June 1,2022 were searched,the number of cases and TCM syndrome types in the literatures were counted,and the rules of TCM syndrome types of lung cancer were discussed.(2)9 of 36 SPF C57BL/6 mice were randomly divided into model group A,traditional Chinese medicine group B,cisplatin group C and combination group D for 14 days after successful modeling.The relative tumor volume,tumor growth rate and tumor inhibition rate were calculated by measuring tumor volume and mass,and the effect of Jiaweifeifu decoction on tumor bearing mice tumor growth was observed.The autophagosomes of tumor tissue cells were observed by transmission electron microscopy.The expressions of LCI/Ⅱ,AKT,p-AKT,m TOR and p-m TOR proteins in tumor tissues were detected by Western blotting.Results: 1.Among the syndrome-types of lung cancer,Qi-yin deficiency(481 cases,17.79%),spleen deficiency phlegm-dampness(476 cases,17.58%)and lung-qi deficiency(277 cases,11.39%)accounted for more than 10%.Most of the syndromes are deficiency syndrome,deficiency syndrome mixed with solid syndrome.According to the distribution of syndrome elements of disease location,the disease location is mainly in lung and spleen.Qi deficiency,Yin deficiency,phlegm,dampness and heat are the key pathological factors.2.(1)Compared with group A,the relative tumor volume in group B,C and D was lower than that in model group,and group D was the lowest(P < 0.001).The relative tumor proliferation rates of group B,Group C and group D were 71.5%,66.57% and 45.33%,respectively.The relative tumor growth rate of group D was the lowest,indicating that TCM combined with cisplatin had the most obvious inhibitory effect on tumor proliferation.Comparison results of tumor weight among all groups: Group B,C and D were lower than group A,and group D was the lowest(P < 0.05).The inhibitory rates of group B,group C and group E were 25%,34% and50%,respectively,indicating that Jiawei Fei Compound had an inhibitory effect on tumor growth.(2)Under electron microscope,autophagosomes/autophagolysosomes were observed in each group,and phagocytic structure was observed in the D group.In group B,the cytoplasm was uniform,the cell membrane was complete,and the edema was light.In group A,C,D,the edema was serious,and some organelles swelled obviously.(3)Western blot results showed that the protein expression of LC3Ⅱ/I in tumor tissues of all groups was higher than that in group A,and the highest was in group D(P < 0.05).Compared with group A,the expression of AKT and m TOR in tumor tissues of groups B,C and D had no significant differences(P > 0.05).The protein expressions of p-AKT and p-m TOR in all groups were lower than those in group A,and the expressions of p-AKT and p-m TOR in group D were the lowest.Conclusion: The most common type of lung cancer is deficiency of Qi and Yin,followed by deficiency of spleen and phlegm and dampness,so the treatment mainly focuses on nourishing qi and Yin.The disease is mainly in the lung and spleen.Qi deficiency,Yin deficiency,phlegm,dampness and heat are the key pathological factors.Jiaweifeifu decoction can improve the general condition of Lewis tumor bearing mice with lung adenocarcinoma,inhibit the tumor growth rate,and enhance the autophagy of tumor cells,which may be related to the down-regulation of AKT/m TOR signaling pathway. |
