| Objective:Comparing the differences of behavioral changes,the striatine dopamine,the Ghrelin and the fabric of intestinal flora on TS model rats under different interventions.Exploring the mechanism of Jiawei xieqing pills in the treatment of TS.Methods:The 40 male SD rats were randomly divided into 10 blank groups and 30 model group,and the model group was prepared by intraperitoneal injection of IDPN to prepare TS animal models,and they were randomly divided into 3 groups:model group,Tiapride group,and Jiawei xieqing pill group.From the date of successful molding,gastric administration is started,the blank group and the model group are given distilled water,and the others groups are given the corresponding liquid,1 time/day.After the 2 weeks of administration,the behavior changes of rats was observed,and the levels of striatine dopamine and serum Ghrelin were detected by ELISA,and 16S rDNA sequencing was used to detect the composition of intestinal contents.Results:(1)After medication,the stereotyped behavior and motor behavior scores of rats in the Jiawei xieqing pill group were significantly reduced(P<0.01);(2)Compared with the blank group,the dopamine content of the striatum in the model group was increased(P<0.05);compared with the model group,the dopamine content of the striatine in the Tiapride group and the Jiawei xieqing pill group was reduced(P<0.05).(3)Compared with the blank group,the content of cerebral intestinal peptide Ghrelin in the model group was increased but not statistically significant(P>0.05);compared with the model group,the content of cerebral intestinal peptide Ghrelin in the Tiapride group was reduced but not statistically significant(P>0.05),and the content of cerebral intestinal peptide Ghrelin in the Jiawei xieqing pill group was reduced(P<0.01).(4)Compared with the blank group,the Ace index and Chaol index of the model group were decreased(P<0.05);compared with the model group,the Ace index and Chao 1 index of the Jiawei xieqing pill group were increased(P<0.05);the Ace index and Chao1 index of the tiapride group were increased,the Shannon index and The Simpson index were decreased,and the Simpson index decreased in the Jiawei xieqing pill group,but none of them were statistically significant(P>0.05).(5)At the gate level,the proportion of Bacteroides in the Jiawei xieqing pill group decreased;At the genus level,the proportion of Lactobacillus species in the Tiapride group and the Jiawei xieqing pill group was increased,and the difference was statistically significant(P<0.05).Conclusion:Jiawei xieqing pills improve abnormal behavior in TS model rats,and their mechanisms may be related to regulating Ghrelin expression and improving intestinal flora. |
