| BackgroundSince the outbreak of the novel coronavirus pneumonia(COVID-19),more than 160 million people have been infected worldwide and more than 5 million people have died,affecting an unprecedented number of people and regions,and having a great impact on countries around the world.The World Health Organization(WHO)says COVID-19 has become a serious threat to human life and global public health security.With the deepening understanding of the evolution of diseases,we have found that the novel coronavirus(SARS-CoV-2)not only affects the respiratory system,but also endangers the gastrointestinal tract and immune system.In the absence of specific treatment drugs,people are opting to vaccinate against the novel coronavirus in order to achieve universal immunity.With the popularization of vaccination,the number of severe cases and deaths of the novel coronavirus has decreased significantly in China.However,due to the strong stealth and infectivity of the novel coronavirus,it is still spreading in a small area in China.Through previous literature,clinical and experimental studies,our team found that intestinal flora can affect the efficacy of vaccines by regulating the immune response and drug metabolism in the host body,and abdominal massage can improve the composition of intestinal flora,regulate the release of inflammatory factors,and enhance the immune function of the body.Therefore,the study on the effect of abdominal massage on the inactivated vaccine of novel coronavirus is of great significance in realizing universal immunity and blocking the transmission of the virus.ObjectiveIn order to confirme that abdominal massage could improve the level of antibody production and the immunogenicity of the vaccine after inoculation;To investigate the effect of abdominal massage on immune regulation of rats injected with COVID-19 vaccine and the mechanism of manipulation;To investigate the effect of abdominal massage on intestinal microorganisms in rats injected with COVID-19 vaccine.MethodsIn this study,immune model rats were manufactured by intramuscular injection of 2 doses of inactivated novel coronavirus vaccine.After modeling,rats in the massage group and vaccine massage group were respectively subjected to abdominal massage manipulation for 8 weeks.At the end of the 8th week,samples were collected from rats in each group to detect the contents of specific antibody IgG,IgA and IgM in peripheral blood.To clarify the effect of abdominal massage on the immune protection of COVID-19 vaccine.Inflammatory factors such as IL-2,IFN-y,IL-4 and IL-5 were detected by ELISA;protein contents of p38MAPK and NF-κBp65 in spleen were detected by WB;intestinal flora of rats was detected by 16S rRNA high-throughput sequencing method.To investigate the mechanism of abdominal massage manipulation on immune regulation in rats inoculated with novel coronavirus vaccine.Results1.The levels of specific antibody IgG,IgA and IgM in peripheral blood of rats were significantly increased after 2 doses of inactivated novel coronavirus vaccine(p<0.05).After 8 weeks of abdominal massage intervention,the levels of specific antibody in peripheral blood of rats with vaccine model showed an increasing trend(p>0.05).2.Compared with normal group,the levels of IL-2 and IFN-y in serum of rats in vaccine group were significantly increased(p<0.05),and the levels of IL-4 and IL-5 were increased(p>0.05).After 8 weeks of abdominal massage intervention,compared with the vaccine group,the serum levels of IL-2,IFN-y,IL-4 and IL-5 in the vaccine model group were increased(p>0.05).3.Compared with normal group,the contents of p38 MAPK,P-P38 MAPK and NF-κB protein in spleen tissue of rats in vaccine group were significantly increased(p<0.05);After 8 weeks of abdominal manipulation,compared with vaccine group,the contents of p38 MAPK,P-P38 MAPK and NF-κB protein in spleen tissue of vaccine massage group showed an increasing trend(p>0.05).4.Analysis of bacterial colony composition:Abdominal massage therapy and novel coronavirus vaccine can regulate the structure of intestinal flora in rats,reducing the ratio of Firmicutes to Bacteroidetes,and increasing the abundance of Bacteroidota,Bacteroidia,Bacteroidales,Oscillospirales,Erysipelaceae,Enterobacteriaceae,Turicibacter and Blautia.Decreased the abundance of Blautia,Lactobacillales,Lachnospirales,Lactobacillaceae,Lachnospiraceae and Alistipes.Conclusion1.Abdominal massage can improve the serum antibody level of rats after vaccination,indicating that abdominal massage can promote the immune response of the body and improve the immune effect.2.Abdominal massage can significantly increase the levels of IL-2,IFN-y and IL-4 in serum of rats,and to a certain extent increase the content of IL-5,thus stimulating better cellular and humoral immunity.3.Abdominal massage can improve the content and phosphorylation level of p38 MAPK and NF-κB p65 in rat spleen tissue,and activate the p38 MAPK/NF-κB signaling pathway.4.Abdominal massage can change the intestinal flora structure of rats inoculated with the novel CoV vaccine,reducing the ratio of Firmicutes to Bacteroidetes,and increasing the abundance of Bacteroidota,Bacteroidia,Bacteroidales,Oscillospirales,Erysipelaceae,Enterobacteriaceae,Turicibacter and Blautia.Decreased the abundance of Blautia,Lactobacillales,Lachnospirales,Lactobacillaceae,Lachnospiraceae and Alistipes. |
