| Objective: In order to investigate the chemical constituents and pharmacological effects of Salvia bowleyana in a more comprehensive way,the chemical constituents of petroleum ether extract and ethyl acetate extract of Salvia bowleyana were isolated and identified systematically,and the antitumor activity of extractum from different polar parts and some pure compounds of Salvia bowleyana were screened.Methods: In this paper,the petroleum ether extract and ethyl acetate extract of Salvia bowleyana were isolated through column chromatography over silica gel,semi-preparative liquid chromatography,D101 macroporous resin column chromatography,reversed-phase ODS column chromatography and Sephadex LH-20 column chromatography and other methods,and the chemical structure identification of the isolated pure compounds by modern spectral analysis techniques and reference to the data reported in the literature.Finally,The antitumor activity of extracts from different polar parts and three pure compounds of Salvia bowleyana was studied by MTT method in vitro.Results:(1)Eleven compounds were isolated and identified from Salvia bowleyana.These compounds are tanshinone IIA(1),cryptotanshinone(2),myristosterol(3),munjistin methy lester(4),nepetoidin B(5),(E,E)-nepetoidin B(6),β-sitosterol(7),ergosterol(8),linolenic acid(9),linoleic acid(10),salpalaestinin(11).Among them,compounds 3,4,6,8,9 and 11 are isolated from Salvia bowleyana for the first time.(2)MTT method was used to investigate the antitumor activity of ethyl acetate extract,nbutanol extract of Salvia bowleyanaand three pure compounds in vitro.Rat glioma cells were used as receptor cells.The results showed that the n-butanol extract of Salvia bowleyana,compounds 1,compounds 5 and compounds 8were screened for cytotoxic activity against Rat glioma cells.Among them,compounds 1 was the best cytotoxic activity against Rat glioma cells with IC50 was0.494 μg/m L.All of four groups were concentration-dependent on the cytotoxic activity of Rat glioma cells.Conclusion: A total of 11 compounds were isolated and identified from the roots of Salvia bowleyana,six of which were isolated from this plant for the first time.the n-butanol extract of Salvia bowleyana,compounds 1,compounds 5 and compounds 8showed an inhibitory effect on rat glioma cells,with a certain dose dependence. |
