Gynura segetum, belonging to the Gynura Genus of Compositae, was firstly recorded in the South Yunnan Materia.As a kind of traditional Chinese materals,the rhizome and root of Gynura segetum (Lour.) Merr. is used to reduce swelling and pain, promote blood circulation and remove blood stasis, arrest bleeding and relieve inflammation, prevent cancer, prevent malaria and cure Bi syndrome, clear heat and detoxicate, etc.. Kinds of biological activities, including anticancer activities, had been found from the Gynura Genus. Researches support that abstraction of Gynura Genus could prevent skin tumor, too. But the anticancer components'material foundation and mechanisms are not very clear yet.Appropriately smashed Gynurae Rhizoma et Radix was extracted by 70% ethanol. The solution was evaporated with a vacuum evaporator and the residue was extracted respectively by petroleum ether, methylene chloride, ethyl acetate, n-Butanol, then got the petroleum ether fraction, methylene chloride fraction, ethyl acetate fraction, n-Butanol fraction and aqueous fraction. The screening of these fractions based on antitumor was carried out on SMMC-7721 and ECA-109 by MTT approach. The results showed that the fraction of methylene chloride and n-Butanol had significant effect on antitumor activities and SMMC-7721 was the sensitive line of the two tested lines.Six compounds from the activity fractions were isolated by various kinds of chromatographies. They were isolated and furtherly identified as Quercetin,β-sitosterol,E-seneciphylline, Senecionine, Seneciphylline,β-daucosterol by spectrum technology such as NMR.An antitumor activity screening was carried out on SMMC-7721 by MTT assay. The assay results showed that E-seneciphylline and Senecio-nine were the activity components among the six substances, whose IC50 were 116.64μg·mL- 1 and 95.95μg·mL-1 respectively. It was deduced that Senecionine's antitumor activity was similar with carboplatin. The antit-umor activity of E-seneciphylline and Senecionine were reported for the first time. Two conclusions can be drawn from the result of the morpho-logical observation on SMMC-7721 by AO assay. First, Senecionine had a good inhibitation on SMMC-7721. Second, Senecionine had significant inhibitory effect on SMMC-7721 in a dose-dependent in vitro. The exp-eriment will be helpful for the further systematic researches of Gynurae Rhizoma et Radix.
