Objective:Based on the data mining of Python programming language,we analyzed the prescription pattern of Professor Liu Hua’s medical cases for primary bronchial lung cancer and extracted the core prescriptions;and explored the mechanism of action of the core prescriptions by using network pharmacology.Through the above study,Professor Liu Hua’s clinical experience in treating primary bronchopulmonary cancer was summarized to guide clinical practice.Methods:1.Data mining research:collect the case data of primary bronchopulmonary cancer patients treated by Professor Liu Hua in the First Affiliated Hospital of Hunan University of Traditional Chinese Medicine in the period from January 1,2020 to June 30,2022,and summarize them into an EXCEL file after standardized processing,use Python programming language software to analyze the high-frequency important and basic properties of drugs in the prescriptions of medical cases,explore the common drug We used the Python programming language software to analyze the high-frequency important and basic properties of drugs in the prescriptions,explore the common drug combinations,refine the core prescriptions,and summarize Professor Liu Hua’s prescription rules for primary bronchopulmonary cancer.2.Network pharmacology study:The core prescriptions from the data mining results were selected for network pharmacology study.Firstly,the active ingredients and corresponding targets of the core prescriptions were collected by TCMSP and Swiss Target Prediction database,and the targets related to primary bronchial lung cancer were obtained by GeneCards,DisGeNET and OMIM databases,and the"active ingredient-disease target" network map of the core prescriptions for primary bronchial lung cancer was constructed by Cytoscape 3.9.1 software.The network of "active ingredient-disease target" was constructed by Cytoscape 3.9.1 software;then the potential PPI was constructed based on STRING database,and the key targets were identified based on the degree value;finally,GO and KEGG enrichment analysis of potential targets were performed by Metascape database to explore the mechanism of action of the core prescription.Results:1.Data mining results:a total of 2183 prescriptions of Chinese medicine tonics were screened,involving a total of 259 Chinese medicines,with a total of 37482 times of drug frequency.The five tastes of the drugs used are,from high to low,sweet,bitter,pungent,light,astringent,and sour;the attributions of the drugs used are,from high to low,lung,spleen,stomach,heart,liver,kidney,small intestine,large intestine,gall bladder,pericardium,and bladder;the efficacy of the drugs used are,from high to low,tonic for deficiency,phlegm,cough,heat,blood circulation,and drainage;27160 groups of drugs were obtained based on the association rule analysis.The most frequently used drug combinations with close association were 27160 groups,among which the highest frequency was 1267 times for the Atractylodes-taxoginseng pair;the core prescription consisted of 10 herbs,including Radix Platycodon,Aster,Radix et Rhizoma,Radix Codonopsis,Atractylodes,Rhodiola,Chrysanthemum,Rhizoma alba,Cyperus,and Cat’s Claw.2.Network pharmacology study:91 active ingredients including quercetin,kaempferol,lignan,spinach sterol,etc.in the core prescription can cross-act with 160 primary bronchial lung cancer targets,and the key targets include TP53,STAT3,AKT1,JUN,TNF,MAPK3,EGFR,IL-6,MAPK1,etc.GO enrichment analysis yielded Biological Process(BP)analysis of 1848,Molecular Function(MF)analysis of 180,Cellular Component(CC)analysis of 96;KEGG pathway enrichment analysis of 196 pathways,which are mainly involved in the regulation of cancer pathway The KEGG pathways were enriched to 196 pathways,including PI3K-Akt pathway,MAPK pathway,AGE-RAGE pathway,and viral infection pathway.Conclusion:1.Professor Liu Hua’s treatment of primary bronchopulmonary cancer is based on benefiting the lung,strengthening the spleen,and harmonizing yin and yang.The treatment is based on pacifying the lung and spleen,regulating lung qi,and eliminating evil and fighting tumors,emphasizing both the symptoms and the root cause,harmonizing yin and yang,as well as using pacifying drugs,attacking and supplementing at the same time,reflecting Professor Liu’s academic thought of "balancing the lung and spleen,supporting the righteousness and fighting tumors".2.Through the analysis of the active ingredients,potential targets and signaling pathways of the core prescription of Eryngium,Aster,Radix et Rhizoma,Atractylodes,Rhodiola,Chrysanthemum,Rhizoma alba,Cichorium,and Cat’s Claw,we predicted that the core prescription inhibits the proliferation,metastasis and invasion of tumor cells mainly by participating in the regulation of cancer pathway,PI3K-Akt pathway,MAPK pathway,AGE-RAGE pathway,and viral infection.The analysis predicted that the core prescription inhibits the proliferation,metastasis and invasion of tumor cells mainly through the regulation of cancer pathway,PI3 K-Akt pathway,MAPK pathway,AGE-RAGE pathway,and viral infection pathway,which laid the foundation for further animal testing or cytological testing to verify its authenticity and effectiveness.
