| Objective:To observe the effect of Yishen Tonglong Decoction on the expression of N-cadherin and vimentin in the nude mouse model of Prostate cancer(Pca).Methods:The PCa subcutaneous implant tumor nude mouse models were established with prostate cancer PC-3 cell suspension,and after successful modeling,they were randomly divided into model groups,low,medium and high dose groups of Yishen Tonglong Decoction,each group of 10,each group was given 0.9%normal saline and low(0.36g/mL),medium(0.72g/mL),high(1.44g/mL)dose of Yishen Tonglong Decoction gastric administration,twice a day for 3 weeks.After the intervention,the general state of the nude rat was observed,the weight and tumor size were measured,and the tumor suppression rate was weighed and calculated.Microscopic observation of morphological differences in nude mouse implant tumors after HE staining;Elisa method detected the levels of the above proteins in the serum of nude mice in each group;PCR detected the mRNA expression of the above proteins in the tissues of nude mice in each group;Western Blot detected the expression of N-cadherin and Vimentin proteins in the tumor tissues of nude mice in various groups.Results:Among them,the serum Vimentin level and the expression level of mRNA of N-cadherin in the dose group of Yishen Tonglong Decoction were not statistically significant compared with the high dose group(p>0.05).1.The tumor inhibition rates in the low,medium and high dose groups of Yishen Tonglong Decoction were 26.67%,42.45%and 66.26%,respectively,and there were statistical differences between the groups(p<0.05);2.HE results showed that compared with the model group,the vacuoles and hyaline-like changes in tumor tissues in the low,medium and high dose Chinese medicine group were significant;3.The results of ELISA showed that compared with the model group,the serum levels of the above proteins in the low,medium and high dose groups of Yishen Tonglong Decoction were reduced(p<0.05),and there was no significant difference in the expression of Vimentin in the serum of nude mice in the high dose group and the medium dose group(p>0.05);4.The PCR results showed that compared with the model group,the expression of mRNA of the above proteins in the low,medium and high-dose groups of Yishen Tonglong Decoction was reduced(p<0.05),and there was no significant difference in the expression of N-cadherin in the nude mouse tumor tissue in the high dose group and the middle dose group of Yi Kidney Tong decoction(p>0.05);5.The results of Western Blot showed that compared with the model group,the expression of N-cadherin and Vimentin in the tumor tissues of the low,medium and high doses of Yishen Tonglong Decoction were significantly reduced,and there were differences between the groups(p<0.05).Conclusion:Yi Kidney Tong Tongyi decoction has a certain inhibitory effect on the expression of N-cadherin and Vimentin proteins in the nude mouse model of PCa subcutaneous implant tumor.The mechanism may be related to the process of epithelial mesenchymal transition(EMT)by regulating the levels of N-cadherin and Vimentin. |
