| Objective:To investigate the effects of Yishen Tonglong Decoction on prostate cancer(PCa)subcutaneous implantation tumor model in nude mice.Nuclear factor E2 related factor 2(Nuclear Factor-erythroid2-related Factor2,Nrf-2)and superoxide dismutase(Super Oxide Dismutase 1,SOD1).Methods:In the groin male BALB/c nude mice inoculated PC-3 cell suspension,establish nude mouse prostate cancer subcutaneous tumor model,after the success of the building were randomly divided into model group,Yishen Tonglong Decoction low,medium and high dose group,model group,normal saline lavage(1 m 1·100g-1·d-1),Experimental group were 0.36g and 0.72g and 1.44g/ml concentration content lavage(1ml·100g-1·d-1),continuous lavage21 d,respectively in the 1d,13d,21 d measuring weight.12h after the last intragastric administration,the nude mice were sacrificed for sampling,and the tumor weight was measured to calculate the tumor inhibition rate.Pathological changes were observed under the HE staining microscope.The contents of Nrf-2 and S OD1 in serum were detected by enzym-linked immunosorbent assay(ELISA),and the protein and mRN A expression levels of Nrf-2 and S OD 1 were detected by Westernblot and RT-PCR.Results:After 21 days of gavage,the body weight of Yishen Tonglong Decoction high-dose group tended to be stable,while the body weight of model group decreased significantly(P<0.05).Compared with model group,the inhibitory rate of tumor in experimental group was increased(P<0.05,P<0.01).HE staining showed that there were different degrees of nuclear lysis in the experimental group.ELISA results showed that Yishen Tonglong Decoction could effectively do wn-regulate the concentrations of Nrf-2 and S OD 1 in serum(P<0.05,P<0.01).The results of WB and RT-PCR showed that Yishen Tonglong Decoction could inhibit the expression of Nrf-2 and SOD1 protein and mRNA(P<0.05,P<0.01).All the above results are concentration-dependent.Conclusion:Yishen Tonglong Decoction can improve the symptoms of nude mice and inhibit the growth of subcutaneous implanted tumors in PC a nude mice,and its mechanism may be related to down-regulating the expression levels of Nrf-2 and S OD 1,promoting the high oxidative stress response of cancer cells,causing cell apoptosis,and then controlling the tumor growth. |
