| Objective: To observe the effects of Simo decoction on intestinal flora,FMO3 and TMAO in rats with heart failure,and to preliminarily explore the possible mechanism of Simo Decoction in improving heart failure from the aspect of intestinal flora.Methods: Forty male SPF-grade SD rats were selected and divided into 2 groups: normal group and model group,with 20 rats in each group.Intraperitoneal injection of isoprenaline was taken for 4 weeks to make a model,and after the completion of the modeling,cardiac ultrasound was used to evaluate whether the modeling was successful,the normal group was randomly divided into control group and control Simo Decoction group,and the model was divided into model group and model Simo Decoction group.And the relevant indicators were tested after 2 weeks of continuous gastric gavage treatment after successful evaluation.Mass spectrometry was used to detect rat plasma TMAO,Western Blot was used to detect rat liver FMO3,ELISA was used to detect rat serum NT-pro BNP,and 16 S r RNA was used to detect rat fecal samples,and the structural characteristics of microbiota between samples were analyzed and compared based on bioinformatics methods.Results: The plasma TMAO content in the model group and the model Simo Decoction group was significantly higher than that in the control group(P<0.05),and the plasma TMAO content in the model Simo Decoction group was less than that in the model group(P<0.05).The liver FMO3 content of rats in the model Simo Decoction group was significantly higher than that in the control group(P<0.05),and the liver FMO3 content in the model Simo Decoction group was smaller than that in the model group(P<0.05),and the serum NT-pro BNP content in the model Simo Decoction group was significantly smaller than that in the model group(P<0.01).The results of 16 S r RNA showed that there was little difference between the control group and the control Simo Decoction group,and there were some differences between the control group and the model group,and the microbiota species between the model group and the model Simo Decoction group.Conclusion: Simo Decoction can reduce the contents of TMAO、 NT-pro BNP and FMO3 in rats with heart failure,and the mechanism may be related to the change of intestinal flora in rats with heart failure. |
