Objective:The pathogenesis of insomnia is closely related to the disorder of 5-HT and Glu/GABA-Gln metabolic loop.This study focuses on 5-HT,Glu,and GABA to investigate the effects of 5-HT,Glu,,GABA content and Glu/GABA ratio,and the behavioral changes of rats with liver depression type insomnia after treatment;explore the curative effect and pathway of electroacupuncture treatment of liver depression type insomnia rats,and provide experimental basis for clinical practice.Materials and methods: Select 48 8-week-old healthy SPF SD rats and feed them adaptively for 3 days.Rats were divided into blank group,model group,western medicine group and electroacupuncture group according to random number table method.A rat model of insomnia due to liver depression was established by intraperitoneal injection of PCPA.During the treatment period,the electroacupuncture group was treated with electroacupuncture,selecting bilateral "Sanyinjiao","Shenmen","Taichong" and "Ganshu" points,the frequency of electroacupuncture was 60 Hz,the intensity was 1 m A,once a day,each time 10 minutes,a total of 6 days;the western medicine group was given diazepam solution 0.9mg/kg,1ml each time,once a day,for 6 consecutive days.Observe the general condition of the rats,body weight changes,elevated maze test,open field test,ELISA to detect the expression of5-HT and GABA in the hypothalamus,and detect the content of Glu in the hypothalamus by measuring glucose.Result:1.Before the experiment,there was no significant difference in the general condition,body weight,elevated maze test and field test of the rats(P>0.05),clustering,nervousness,anxiety,aggressiveness increased,activity decreased,food and water intake decreased,The weight gain of the model group was significantly lower than that of the blank group(P<0.05),the ratio of the number of times of entering the open arm to the time of entering the open arm in the elevated maze test was significantly reduced(P<0.05),and the number of fecal particles was significantly increased(P<0.05).2.After electroacupuncture and western medicine treatment,the softness and luster of the hair of the rats increased,the sense of tension and anxiety was improved,the stool was formed,and the diet and drinking water were improved.The aggression decreased,and the mood recovered and stabilized;the weight of the rats in the above two groups was significantly higher than that of the model group(P<0.05),and the weight of the rats in the electroacupuncture group was significantly higher than that of the western medicine group(P<0.05).3.Compared with the rats in the blank group,the ratio of the number of times of entering the open arm in the ascending maze test to the entry time of the model group was significantly reduced,the score of the open field test was significantly reduced,and the number of fecal particles was significantly increased(P<0.05);electroacupuncture and After western medicine treatment,compared with the model group,the ratio of the number of times the rats entered the open arm and the ratio of the entry time increased significantly(P<0.05),and there was no significant difference between the two groups(P>0.05);the rats in the two treatment groups Compared with the model group,the score of the open field test was significantly increased,and the number of fecal particles was significantly reduced(P<0.05),and there was no significant difference between the two groups(P>0.05).4.Compared with the blank group,the 5-HT content in the hypothalamus of the rats in the model group was significantly lower(P<0.05);after electroacupuncture and western medicine treatment,the 5-HT content in the hypothalamus of the rats was significantly higher than that in the model group(P<0.05),the improvement in the electroacupuncture group was significantly higher than that in the western medicine group(P<0.05).5.Compared with the blank group,the expression of GABA in the hypothalamus of rats in the model group was significantly reduced(P<0.05);after treatment,the expression of GABA in the hypothalamus of rats in the electroacupuncture group and western medicine group was significantly higher than that in the model group(P<0.05)),the expression of GABA in the electroacupuncture group was significantly higher than that in the western medicine group(P<0.05).6.Compared with the blank group,the expression of Glu in the hypothalamus of rats in the model group was significantly increased(P<0.05);after electroacupuncture and western medicine treatment,the expression of Glu in the hypothalamus of rats in the two groups was significantly lower than that in the model group(P<0.05).7.Compared with the blank group,the Glu/GABA ratio of the rats in the model group was significantly increased(P<0.05);after electroacupuncture and western medicine treatment,the Glu/GABA ratio of the two groups of rats was significantly lower than that of the model group(P<0.05),the electroacupuncture group decreased more significantly than the western medicine group(P<0.05).Conclusion:1.Electroacupuncture at "Sanyinjiao","Shenmen","Taichong" and "Ganshu" points can effectively improve the general condition,body weight,elevated maze test and open field test of insomnia rats;increase the expression of 5-HT and GABA in the hypothalamus of insomnia rats,reduce the expression of Glu in the hypothalamus,and down-regulate the ratio of Glu/GABA.Although the Western medicine group can also improve the expression of 5-HT,GABA and Glu,the effect The strength is not as good as that of the electroacupuncture group.2.The therapeutic effect of electroacupuncture at "Sanyinjiao","Shenmen","Taichong" and "Ganshu" on rats with liver depression and insomnia may be through Glu/GABA-The Gln metabolic circuit improves the expression of 5-HT,GABA and Glu,thereby repairing sleep disorders in rats and relieving the anxiety state of rats.
