| Purpose: Based on the NF-κB signalling pathway,the expression of NF-κBp65,p-CREB and AQP5 in rats with allergic rhinitis was observed by using Forskolin(c AMP activator),PDTC(NF-κB inhibitor),H89(PKB inhibitor),loratadine and large,medium and small doses of Xiaoqinglong Decoction,Ephedra,Xixin and Fuzi Decoction combined with Yupingfeng Powder.The expression of NF-κBp65,p-CREB and AQP5 in the combined formula of Xiaoqinglong Decoction,Ephedra,Xixin and Fuzi Decoction and Yupingfeng Powder for the treatment of allergic rhinitis was investigated in depth.Material and Method: Ninety clean-grade healthy male Wistar rats were randomly divided into 9 groups according to body weight.That is blank control group,the model control group,the Forskolin intervention group,the H89 intervention group,the PDTC intervention group,the loratadine group,Xiaoqinglong Decoction,Ephedra and Fuzi Decoction combined with Yupingfeng Powder small dose group,Xiaoqinglong,Ephedra Ma Fuzi Decoction combined with Yupingfeng Powder medium-dose group,10 rats in each group.The AR rat model was made using a combination of systemic sensitization and local attack using an ovalbumin suspension for all groups except the blank control group.The blank control group and model control group were given saline gavage at 1 ml/100 g;PDTC intervention group was given PDTC at 50 mg/kg/d intraperitoneally;H89 intervention group was given H89 at 5 mg/kg/d intraperitoneally;Forskolin intervention group was given Forskolin at 5 mg/kg/d intraperitoneally;loratadine treatment group was given loratadine solution at 1 mg/kg/d.The loratadine solution was given by gavage;the decoction of the tested drug was given by gavage at 7.1 ml/kg(equivalent to 7.1 g of raw drug),25.0 ml/kg(equivalent to 25.0 g of raw drug)and 44.2 ml/kg(equivalent to 44.2 g of raw drug)in the small,medium and large dose groups of Xiaoqinglong Decoction,Ephedra,Xixin and Fuzi Decoction and Yupingfeng Powder respectively.After the last nasal drip,observe the performance of scratching,sneezing and runny symptoms in each group of rats.Then the rats were anaesthetized and executed,and the nasal mucosa of each group was taken and the expression of p-CREB,AQP5 and NF-κBp65 was detected by Western blotting,and the results were analysed statistically.Results:1.The behavioral characteristics of rats after treatment were compared: Forskolin,PDTC and loratadine groups all improved the symptoms of AR rats(p<0.05).H89 intervention group failed to improve the symptoms of AR rats.The combination of Xiaoqinglong Decoction,Ephedra,Xixin and Fuzi Decoction and Yupingfeng Powder could improve the symptoms of AR rats(p<0.05),and the effect was more obvious in medium and large doses(p<0.05).2.NF-κBp65 Western blotting results: the expression of NF-κBp65 in the model group was significantly higher than that in the blank control group(p<0.05).The expression of NF-κBp65 in the Forskolin intervention group was lower than that in the model group.The expression of NF-κBp65 in the PDTC intervention group and loratadine group was significantly lower than that in the model group(p<0.05).The expression of NF-κBp65 in the H89 intervention group was significantly higher compared with that in the model group(p<0.05).The expression of NF-κBp65 was significantly lower in the Xiaoqinglong Decoction,Ephedra,Xixin and Fuzi Decoction and Yupingfeng decoction combined formula compared with the model group(p<0.05),and the reduction was more significant in the medium and high dose groups(p<0.05).3.The expression of p-CREB in the model group was significantly lower than that of the blank control group(p<0.05).p-CREB expression in the Forskolin,PDTC,and loratadine groups was significantly higher compared with the model group(p<0.05).p-CREB expression in the H89 intervention group was lower than that of the model group(p<0.05).The combination of Xiaoqinglong Decoction,Ephedra,Xixin Hsio and Fuzi Decoction,and Yupingfeng Powder could increase the p-CREB expression(p<0.05),and the increase was more significant in medium and large doses(p<0.05).4.AQP5 Western blotting results: AQP5 expression in the model group was significantly lower than that in the blank control group(p<0.05);AQP5 expression in the Forskolin intervention group,PDTC intervention group and loratadine group was significantly higher compared with the model group(p<0.05);AQP5 expression in the H89 intervention group was lower than that in the model group.The combination of Xiaoqinglong Decoction,Ephedra,Xixin and Fuzi Decoction and Yupingfeng powder could increase AQP5 expression(p<0.05),and the increase was more significant in medium and large doses(p<0.05).Conclusion: AQP5 expression could be increased by inhibiting the activation of NF-κB signaling pathway by using Xiaoqinglong Decoction,Ephedra,Xixin and Fuzi Decoction and Yupingfeng decoction,thus improving the scratching,sneezing and runny symptoms of AR rats,and the improvement was more significant in medium and large doses. |
