Experimental Study On Anti-allergic Effect Of Mahuang Fuzi Xixin Decoction Based On Allergic Rhinitis

Objective: To verify the anti-allergic effect of Mahuang Fuzi Xixin description,explore its active mechanism,and discuss the effect of interference on allergy,by observing its effect on behavioral characteristics,IgE and histamine,the structure of nasal mucosa and lung tissues in guinea pig models of allergic rhinitis.Methods: We chose 20 male and 20 female 2-3 months regular grade guinea pigs as the research object.Guinea pigs were randomly divided into four groups: blank group,model group,Chinese medicine group and chlorpheniramine group,10 in each.The allergic rhinitis model in guinea pigs were begun to copy after adaptability of feeding for 1 week.Except for the blank group giving the same amount of normal saline in accordance with the following methods,each of the guinea pigs in other groups were given intraperitoneal injection of mixed suspension,which is configured with ovalbumin and aluminum hydroxide,1ml/time,once every other day for 7 times,as well as intranasal ovalbumin solution for partial stimulation,0.05ml/time/side,once a day for 7 times.After the successful construction of the models,the Chinese medicine and chlorpheniramine groups respectively treated with Mahuang Fuzi Xixin Decoction(1mg/ml)and chlorpheniramine(0.2mg/ml)by intragastric administration,the other two groups with equal saline,whose dosages were converted for guinea pigs,once a day and sustained for 10 times.Observing behavioral changes in every guinea pig after 30 minutes of the last medication,anesthetizing them by injecting with 10% chloral hydrate,collecting blood after their death,making nasal mucosa and lung tissues into specimens and observing their changes in pathological morphology,detecting content of IgE and histamine in serum with ELISA.Results: 1 Effect on AR guinea pigs’ ethology: The guinea pigs in blank group occasionally catch nose,sneeze,but there is no symptom of flowing clear nasal mucus.The model group has evident phenomenons mentioned above,and the same symptoms of guinea pigs in Chinese medicine and chlorpheniramine groups can be relieved.2 Effect on IgE and histamine in serum: Mahuang Fuzi Xixin decoction and chlorpheniramine may reduce content of IgE and histamine in serum(P<0.05),the effect of chlorpheniramine in decreasing the content of IgE is better than Chinese medicine(P<0.05),but there is no significant differences between their therapeutic function in decreasing the content of HIS(P>0.05).3 Effect of pathological morphology on guinea pig models of AR:Nasal mucosa tissue: The blank group has intact nasal epithelial structure,and there are no conspicuous changes in glands and blood vessels,the inflammatory cells infiltration are occasional.Compared with the blank group,guinea pigs in model group show a large number of inflammatory cells aggregation and infiltration,along with nasal epithelial hyperplasia,cilia breakdown,goblet cells increasing,glands proliferation and edema associated with a small amount of secretions,blood vessels congestion and so on.Morphology changes of the other two groups are much improved,comparing with the model group,only local epithelial hyperplasia,inflammatory cells decreasing,glands mild proliferation,and the repairing functions of Mahuang Fuzi Xixin decoction and chlorpheniramine are indifference,but there are still discrepancies compared with blank group.Lung tissue: The blank group has intact structure and normal morphology,and occasional inflammatory cell infiltration,mainly lymphocyte.Compared with the blank group,model group present alveolus structural damage,inflammatory cell infiltration,alveolar spaces congestion and edema,pulmonary interstitial thickening.Chinese medicine and chlorpheniramine can significantly reduce the above-described morphological changes,and the two groups are indifference.Conclusion: 1 Mahuang Fuzi Xixin Decoction can relieve the symptoms of catching nose,sneezing and flowing clear nasal mucus in guinea pig models of allergic rhinitis.2 Reducing the concentrations of IgE and histamine in guinea pigs’ serum.3 Improving the structure of nasal mucosa and lung tissue significantly.Showing that this therapy has a positive effect in protecting and restoring the above pathological tissues.
