Objective: Previous studies have demonstrated that miR-32-5p is abnormally expressed in tumors,and plays the dual role of cancer promotion and tumor suppressor genes Multiple studies have confirmed that miR-32-5p is down-regulated in non-small cell lung cancer tissues and cells,plays a role as a tumor suppressor gene,it plays an important role in regulating the proliferation,invasion and migration of lung cancer cells.This study was mainly to investigate the effects of miR-32-5p on non-small cell lung cancer,study its effects on proliferation,migration,and invasion of non-small cell lung cancer A549 cells,and verify its relationship with PI3K/AKT signaling pathway.Methods: The expression of miR-32-5p in human non-small cell lung cancer cell lines A549,H1299,H1975 and normal bronchial epithelial cells 16 HBE were determined by qRT-PCR in this study.A549 cells were transfected with miR-32-5p mimic(miR-32-5p group)and miR-32-5p negative control(NC group),the proliferation of A549 cells was detected by CCK8 method,Western Blot was used to detect the expression levels of migration-related proteins MMP2,MMP9 and PI3K/AKT signaling pathway.Results: Compared with normal bronchial epithelial cells 16 HBE,the expression of miR-32-5p decreased in all non-small cell lung cancer cell lines.Compared with the negative control group,the expression of miR-32-5p was up-regulated after transfection with miR-32-5p mimics,which inhibited the proliferation,migration and invasion of A549 cells.The levels of key protein molecules PI3 K,p-PI3 K,AKT and p-AKT in the PI3K/AKT signaling pathway are down-regulated.Conclusion: Overexpression of miR-32-5p can inhibit the proliferation,migration and invasion of A549 cells,which may be mediated by the regulation of PI3K/AKT signaling pathway. |
