| Objective:To observe the effects of Tiandi Zhuangqi Decoction on learning and memory ability and hippocampal tissue SIRT1,NF-κB and IκBαin AD model rats.Methods:1.Seventy 8 week-old male SD rats were randomly divided into blank group,sham operation group,model group,low-dose treatment group,medium-dose treatment group,high-dose treatment group and western medicine treatment group,with 10 rats in each group.2.The rat model of AD was made by injecting Aβ1-42 into the hippocampus of both sides using stereostereoscope.3.Morris water maze was used to observe the learning and memory of AD model rats.4.Morphological changes of hippocampal neurons in AD model rats were observed by HE staining.5.The expression levels of SIRT1,NF-κB and IκBαin rat hippocampus were detected by Western Blot.Results:1.Effects of Tiandi Zhuangqi Decoction on learning and memory ability of AD model rats:The water maze results showed that compared with model group,the target quadrant swimming time of rats in Aβ+Tianduangqi decoction high-dose and medium-dose groups was significantly prolonged(P<0.05),and there was no statistical significance compared with western medicine group(P>0.05),indicating that Tianduangqi decoction can improve the learning and memory ability of Alzheimer’s disease rats.2.Morphological changes of hippocampal neurons in AD model rats induced by Tiandi Zhuangqi Decoction:In the blank group and the sham group,the neuronal cells were full in shape,regular in structure,abundant in number,compact and neat in arrangement,with clear boundaries,distinct layers,uniform staining,clear,round-like nuclei,and abundant cytoplasm.There was no significant difference between the two groups,indicating that the surgical injury did not cause any histomorphologic changes in the hippocampus.Compared with the sham operation group,the hippocampal neurons in the model group were significantly changed in shape,irregular structure,reduced in size,nucleolar shrinkage,dark staining,turbidity in the cytoplasm,a large number of neurons died,and the number of normal neurons decreased significantly.Compared with the model group,the number of neuron cells in the Chinese medicine treatment group and the Western medicine treatment group was relatively increased,the cell arrangement was relatively neat,the distribution of cells was more uniform,and the intracellular structure was clearer and normal.3.Effect of Tiandi Zhuangqi Decoction on the expression levels of SIRT1,NF-κB and IκBαin rat hippocampus:The results of Wb detection showed that compared with model group,the expression levels of STRT1 and IκBαprotein in Aβ+Tiandi Zhuangqi decoction high and medium dose groups were significantly increased(P<0.05),while the expression levels of NF-κB protein were significantly decreased(P<0.05).The expression level of STRT1 protein in Aβ+Tiandi Zhuangqi decoction high-dose group was significantly higher than that in Aβ+Tiandi Zhuangqi decoction medium-dose group,the difference was statistically significant(P<0.05).Conclusion:1.Tiandi Zhuangqi decoction can improve the learning and memory ability of AD model rats;2.Tiandi Zhuangqi Decoction can improve the morphological changes of hippocampal neurons and inhibit the apoptosis of hippocampal neurons in AD model rats;3.Tiandi Zhuangqi Decoction can inhibit hippocampal neuronal inflammation in AD model rats,and its mechanism may be as follows:By increasing the expression of SIRT1 and related factors,Tiandi Zhuan Qi Decoction can activate the SIRT1/NF-κB signaling pathway,inhibit the expression of NF-κB protein,and increase the expression of IκBα,the self-inhibitor of NF-κB protein,further reduce the expression of NF-κB protein,reduce the inflammatory response of neurons,so as to protect neurons and improve the learning and memory ability of rats.4.Tiandi Zhuangqi Decoction has certain prevention and treatment ability on model rats of Alzheimer’s disease,suggesting that the treatment of Alzheimer’s disease by tonifying kidney,eliminating phlegm and promoting blood circulation has certain feasibility,and is worth further exploration. |
