| Objective:Study the effect of Jiawei Duhuo Jisheng Mixture on bone metabolism markers in rabbit knee osteoarthritis model,and elucidate the mechanism of action of Jiawei Duhuo Jisheng Mixture in treating knee osteoarthritis from the perspective of bone metabolism.Methods:32 female New Zealand white rabbits were randomly divided into 4 groups:blank group,model group,traditional Chinese medicine group,and positive control group,with 8 rabbits in each group.After deep anesthesia,the model group,the traditional Chinese medicine group,and the positive control group were injected with papain into the knee joint cavity once every three days,a total of three times.During the modeling process,the rabbits were placed in an open field at 10 a.m.and 4 p.m.every day to drive away,with a total of forced activity for 30 minutes.The blank group was injected with an equal amount of 0.9%sodium chloride solution,and the other operations were the same as those of other groups.After the completion of modeling,the positive control group was given glucosamine sulfate suspension by gavage,the traditional Chinese medicine group was given an equal amount of Duhuo Jisheng Mixture solution by gavage,and the blank group and model group were given an equal amount of physiological saline by gavage for a total of 8 weeks.After gavage,the general situation of each group was observed,and blood was taken from the ear vein for centrifugation and preservation.The serum levels of CTX-Ⅰ,MMP-3,and TRACP-5b were detected by enzyme linked immunosorbent assay.Results:1.General situation:There is no significant difference between the blank group and the New Zealand white rabbits before modeling.The model group,the Chinese medicine group,and the positive control group,the New Zealand white rabbits,exhibit mental fatigue,lack of activity,and slight loss of appetite.Looking at the knee joint,it is found that the right knee joint is significantly swollen compared to the left,with elevated skin temperature,and local cyanosis.Passive activity of its right knee joint can induce pain reflex.After 8 weeks of gavage,the Chinese medicine group and the positive control group,compared with the model group,have an appetite,Mental state and swelling degree of knee joint have improved.2.Serum CTX-Ⅰ level:The CTX-Ⅰ level in the model group was significantly higher than that in the blank group(P<0.05);Compared with the model group,the level of CTX-Ⅰ in the traditional Chinese medicine group and the positive control group decreased(P<0.05),while there was no statistically significant difference between the traditional Chinese medicine group and the positive control group(P>0.05).3.Serum TRACP-5b level:The level of TRACP-5b in the model group was significantly higher than that in the blank group(P<0.05);Compared with the model group,the level of CTX-Ⅰ in the traditional Chinese medicine group and the positive control group decreased(P<0.05),while there was no statistically significant difference between the traditional Chinese medicine group and the positive control group(P>0.05).4.The level of MMP-3 in serum showed that the level of MMP-3 in the model group was significantly higher than that in the blank group(P<0.05);Compared with the positive control group and the model group,the level of MMP-3 in the traditional Chinese medicine group decreased(P<0.05),while there was no statistically significant difference between the traditional Chinese medicine group and the positive control group(P>0.05).Conclusion:The treatment of knee Osteoarthritis with Jiawei Duhuo Jisheng Mixture may be achieved by regulating the level of bone metabolic markers. |
