Objective: In this study,TLR4/NF-κB signaling pathway mediating low-grade inflammation of gastric mucosa was taken as the entry point,and the intervention mechanism of electroacupuncture at "Zusanli" and "Taichong" points on functional dyspepsia rats was discussed by observing the changes of gastrointestinal momodynamics,TLR4/NF-κB signaling pathway related genes,inflammatory factors content and gastric mucosal inflammatory response.To provide experimental basis for acupuncture and moxibustion clinic.Materials and methods: Forty-eight SPF male and female SD rats of 10 days of age were selected and randomly divided into normal group,model group,electroacupuncture group and non-meridian and non-acupoint group,with 12 rats in each group.In addition to the normal group,the other three groups were treated with iodoacetamide gavage combined with tail clip and irregular diet to prepare compound FD rat models at 10 days of age.The rats in the electroacupuncture group received acupuncture intervention at both "Zusanli" and "Taichong" points by electroacupuncture.The waveform was selected as dense wave,the intensity was 0.5m A,the frequency was 2 Hz/50 Hz,20 minutes a day,and the intervention lasted for 14 days.In the non-meridian and non-acupoint group,both sides of the electroacupuncture "non-meridian and non-acupoint" were treated with the same intervention method as the electroacupuncture group.During the experiment,the general conditions of the rats were observed and their weight was recorded.During the sampling period,6 rats in each group were randomly captured to detect the gastric emptation rate and small intestine propulsion rate.HE observed the pathological morphologic changes of gastric tissue in rats.The expression of interleukin-6(IL-6)in gastric tissues of rats was determined by enzyme-linked immunosorbent assay(ELISA).The protein and m RNA expression of TLR4/NF-κB pathway were detected by immunohistochemistry(IHC)and fluorescence quantitative PCR.Results:1.Before the experiment,there was no difference in the general condition and body weight of each group(P > 0.05),after modeling,compared with the normal group,the mental state of the other three groups was poor,the hair was disordered and lustrous,loose stool was loose,the range of motion was decreased,and the body weight was decreased(P < 0.01);After electroacupuncture intervention,the mental state of the rats in the electroacupuncture group improved,the hair was smooth,the feces was granular,the diet and water intake gradually increased,and the body weight of the rats in the electroacupuncture group increased significantly compared with the model group and the non-acupoint group(P < 0.01).2.HE staining showed a small amount of inflammatory cell infiltration in the gastric mucosa of model group and non-transacupoint group under the microscope(X200),and no obvious abnormalities in the remaining groups.After dissection,the morphology and appearance of gastric tissue were basically normal,and no obvious changes such as congestion,necrosis and ulceration were observed in the naked eye.3.Compared with the normal group,the gastric emptance rate and intestinal propulsion rate of FD rats in model group were significantly decreased(P < 0.01);The gastric emptying rate and intestinal propulsion rate of FD rats in the electroacupuncture group were significantly increased compared with the model group and the non-acupoint group(P <0.01).4.Compared with normal group,the protein expression of TLR4,NF-κBp65,NF-κBp-p65 and the content of IL-6 in gastric tissue of FD rats in model group were increased(P < 0.05),increased expression of TLR4 and NF-κBP65 mrna in gastric tissues(P <0.05);After the electroacupuncture intervention,the protein expression of TLR4,NF-κBp65,NF-κBp-p65 and the content of IL-6 in gastric tissue of rats in the electroacupuncture group were decreased compared with that in the model group(P < 0.05),decreased the expression of TLR4 and NF-κBP65 mrna in gastric tissues(P < 0.05);Compared with the non-acupoint group,the protein expression and IL-6 content of TLR4,NF-κBp65 and NF-κBp-p65 in the gastric tissue of FD rats in the electroacupuncture group were decreased,and the mrna expression of TLR4 and NF-κBP65 in the gastric tissue were decreased(P < 0.05).Conclusion:1.Electroacupuncture at "Zusanli" and "Taichong" points can effectively improve gastrointestinal motility in FD rats,relieve low-grade gastric mucosal inflammation in FD rats,and have a certain intervention effect on FD rats.2.Electroacupuncture at "Zusanli" and "Taichong" points can down-regulate the expression of protein and m RNA related to TLR4/NF-κBp65 signaling pathway and reduce the content of downstream inflammatory factor IL-6,suggesting that electroacupuncture at "Zusanli" and "Taichong" points can improve low-grade gastric mucosal inflammation in FD rats through TLR4/NF-κB signaling pathway.Play a role in the treatment of FD.
