It is investigative and important subject in current medicine field for separating the active composition in plant, the high pure active composition from the plant can be used as cure for many diseases efficiently. The current pharmacological researches have expressed that the ursolic acid is a treatment medicine in high blood fat, diabetes, heart brain blood vessel disease, and cancer with prospect. The current researches concentrate primarily in the pharmacological aspects and have obtained the important results. However, the research of the separation and purification of ursolic acid for the industrialization is less. For making up this shortage, this discourse has studied the isolation of ursolic acid from Ilex kudingcha C. J. Tseng with united technique of ultrafiltration, chemical concentration and macro porous resin adsorption in order to get the short process of high recovery yield and high purity based on industrialization. The research expressed that the design of process route is reasonable, the method is proper and the purity and recovery yield of product come to higher level.1. The content of ursolic acid from Ilex kudingcha C. J. Tseng was examined by HPLC for the first time. Comparing with literature data, the content of ursolic acid from Ilex kudingcha C. J. Tseng was the highest in the studied plants, reached up to 1.44%. Ilex kudingcha C. J. Tseng is a kind of green plant very suitable to extracting ursolic acid, which indicated a high guality medicine resource was exploited for the separation and purification of ursolic acid;2. When ursolic acid was analyzed according to the current methods, the chromatographic peak expanded, meanwhile with the drift of baseline. Aiming at these problems, the acidity material NaH2PO4 and the alkaline material diethylenceamide were added to the flow phase for the first time so that the acidity and alkaline property of flow phase was regulated effectively. Ursolic acid can get the complete separation with other materials in 20 minutes. The baseline is stable and the peak form is good. A kind of special regulate diethylenceamide was found out for the analysis of ursolic acid in order to increase analysis speed and accurate degree;ABSTRACT3. Ursolic acid was leached with the ethanol solvent for its low poison and friendly environment. Leach time affected leaching rate very obviously by orthogonal design. The best process conditions was that 0.320.63 mm of Ilex kudingcha C. J. Tseng powder was leached with 90% alcohol by reflux extraction for twice each for 2.5h at 1 I 14 of Ilex kudingcha C. J. Tseng mass to solvent volume at 85 癈. The experiment result was satisfied when laboratory experiment was scaled up for 220 ~ 250 times, which indicated good enlargement function. Leaching dynamics research showed that leaching process of ursolic acid was governed by pore diffusion.4. The ultrafiltration technique was used to purify leaching liquor of Ilex kudingcha C. J. Tseng for the first time. The pigment, big molecule matter and solid could be eliminated with it effectively. The permeation rate of ursolic acid was 99.2% about poly(ether sulfone)(PES) membrane of trapped molecular weight 10000 chosen, the best operation pressure was 100 ~300 kPa, the optimal concentration of ursolic acid was 0.55 mg/ ml in material solution. The cleaning experiment of membrane discovered that the main component of membrane fouling can be divided into two parts, one could be dissolved in 1% NaOH, the other in 75% alcohol. The membrane flux recovered to 96% of the new membrane when the membrane was cleaned by these two kinds of solution alternately.5. Because of the difficult filtration and low recovery yield of traditional procedure, the concentration process of ursolic acid in the alcohol leaching liquor was investigated for the first time. Ursolic acid was separated availably by utilizing the pH difference of various matters existing in leaching liquor, the emulsion phenomenon of precipitation process was avoided so that the recovery yield of ursolic acid was increased and...
