With broad pharmacological action, the ginkgo biloba L. as Chinese traditional medicine has been applied with long history. The important active ingredients include flavonoids and ginkgolides. Pharmacological researches demonstrated that the pharmacologic effects of flavonoids and ginkgolides are eliminating oxygen-derived free radicals, delaying aging, anti-inflammation, relieving ache, anti-bacteria and relaxing coronary vessels and so on. The focus of recent researches is how to make the extraction and separation process of the active ingredients better from ginkgo biloba L. leaves.In this paper, water and alcohol as extracting solvent were used to extract gingko flavonoids. Then, the technical optimizing of different factors of extraction was discussed in this work. The outstanding factors of extraction progress were the matching of feedstock and solvent, alcohol concentration and extraction temperature with the orthogonal test. The method of the single factor control test was utilized to optimize the important influence factors, as follow: the matching of feedstock and solvent 1:10, alcohol concentration 70%, extraction temperature 70℃, granularity 4080, the time of extracting 2h. The ratio of extraction was up to 86.5%.The analysis method was studied in the paper: Al(NO3)3 is selected as the only characteristic reactant with flavonoids in the extraction solvent. Only has characteristic reaction with flavonoids in the extraction solvent. The spectrophotometry was used for determining the flavonoids in the extractive of ginkgo biloba L. leaves. The wave length of analysis is 415nm. The concentration and the absorbance have good linear relationship within the concentration range of 6.32×10-4-0.476mg/ml. The method was simple, rapid and accurate.The separation and purity of flavonoids in the paper employed the absorption with D101-macro-porous adsorption resin. Its absorption trait was discussed by the method of the Static Adsorption and the Dynamic Adsorption. The adsorption isotherm of different temperature was gained in 10℃, 25℃, 40℃ by static adsorption. The result showed that the adsorbed amount of flavonoids increased with the raise of temperature, the adsorbed amount increased more from 10℃ to 25℃ than from 25℃ to 40℃. Therefore, the...
