Due to the excellent biological diversity, chemodiversity, and bioactive diversity, alkaloids are paid attention very much all over the world. The study on the rapid, sensitive, reliable, qualitative and quantitative analysis method of alkaloids in complicated matrices is continuously popular. In the present paper, the separation and analysis methods of different kinds of alkaloids with high performance liquid chromatography coupled with mass spectrometry, gas chromatography coupled with mass spectrometry,"green"chromatography, and preparative high performance liquid chromatograhy have been systematically studied. The details are summarized as follows:1. Aporphine alkaloids, N-nornuciferine, O-nornuciferine, nuciferine, and roemerine, have been isolated and purified from the leave of Nelumbo nucifera Gaertn using silica gel column chromatography and preparative high performance liqud chromatography. The structures of above four alkaloids have been confirmed by analyzing their spectra of UV, IR, ESI-MS, 1H-NMR, and 13C-NMR. With N-nornuciferine, O-nornuciferine, nuciferine, and roemerine as target compounds, high-performance liquid chromatography-photodiode array detection-electrospray mass spectrometry and gas chromatograpgy-mass spectrometry have been developed for the simultaneous determination of aporphine alkaloids. And the proposed methods have been applied successfully to the determinations of N-nornuciferine, O-nornuciferine, nuciferine, and roemerine in leaves of Nelumbo nucifera GAERTN and health foods.2.α-Naphthaphenanthridine alkaloids, sanguinarine and chelerythrine, have been isolated and purified for the first time from the fruit of Macleaya cordata (Willd) R. Br. using macroporous adsorption resin. The structures of above two alkaloids have been confirmed by analyzing their spectra of UV, IR, ESI-MS, 1H-NMR, and 13C-NMR. With sanguinarine, chelerythrine, protopine, and allocrytopine as target compounds, the isocratic high-performance liquid chromatographic method coupled with electrospray mass spectrometry has been developed to determine simultaneouslyα-naphthaphenanthridine and protopine alkaloids. The method is employed successfully to determine protopine, allocryptopine, sanguinarine and chelerythrine in fruits of Macleaya cordata (Willd) R. Br., and sanguinarine and chelerythrine in the exogenous contaminated honey.3. A high-performace liquid chromatographic method coupled with electrospray mass spectromety has been developed to simultaneously analyse protoberberine, indolequinoline and quinolone alkaloids.
