High-performance liquid chromatography combined with electrospray ion trap mass spectrometry, due to its high separation efficiency and great capability in on-line structural characterization, has become one of the most powerful techniques in pharmaceutical analysis. Using this technique, we investigated the components of alkaloids in natural product of Piper longum L., and impurities in bulk drugs of lisinopril and ketoprofen.The characteristic fragmentation patterns of alkaloids from natural plant of piper longum L. were discussed, and a rapid and sensitive method was developed for the characterization and identification of alkaloids in piper longum L. Within this study. 27 alkaloids were rapidly identified, of which 10 were first found in this plant and 2 were new compounds.The fragmentation mechanism of lisinopril and two related compounds were investigated, and a sensitive and exact method was developed for identification of trace-level impurity in bulk drug of lisinopril. By using this technique, a novel impurity in bulk drug of lisinopril was successfully identified.Based on the comparison of the fragmentation patterns between ketoprofen and impurities, four pairs of trace-level impurities in bulk drug of ketoprofen were rapidly and successfully characterized, and their fragmentation patterns were also discussed.In addition, the principle of LC/MS technique and its extensive application in pharmaceutical analysis was reviewed in detail. |
