Study On Stress Resistance Of Cerasus Humilis(Bge)Sok And Genetic Variation Of Agriculture Characteristics As Well As Molecular Marker Among The Resources

In this study, the materials were eight resources of Cerasus humilis(Bge)Sok from Beijing forest and gardening Bureau of seeds and seedlings station, whose plants were treated by salt and drought stress and some physiological index of the plants were measured, such as percentage of electrolyte exosmosis, chlorophyll content, MDA content, POD and SOD activity, proline content and so on, it the first time to study the physiological machenism of Cerasus humilises against the stress, what will give useful information for selecting good resource and plants with strong salt resistance and drought tolerance. Besides, the growth, leaf performance, and soluble sugar and soluble proten in the leave from 8 resources of Cerasus humilises were also firstly measured and analyzed, as well as the ISSR molecular marker was applied to study genetic diversity of the resources, the purpose was to systematically investigate genetic differetiation, genetic correlation and genetic construction of different resource in molecular level, providing theoretical basis for resource division.The results were as follows:Two years old potted plants --- 2 resource of Cerasus humilises, Nongda No.3 and Nongda No.4 were irrigated with different concentration of NaHCO3 solutions and mensurated physiological indexes of leaves. The results indicated that the change of chlorophylla content in two clone of Cerasus humilis of leaves both submited firstly not clearly then decreased, analysis of variance were significant difference among different disposals; the change of chlorophyll a/b were calmly in the whole stress period; the change of proline content submited firstly not clearly then upregulated; the changes of electrolyte exosmosis percentage, MDA content and POD activity of Nongda No.3 were not clearly firstly then increased and then decreased, and analysis of variance showed there were no significant difference among different disposals, the change of SOD activity in the middle and low concentration were not clear, the chang in high was clear, and analysis of variance showed there were significant difference among different disposals, the change of Nongda No.4 electrolyte exosmosis percentage, SOD activity, POD activity and MDA content submited firstly increased then decreased, after salt stress relieved, exosmosis percentage, SOD activity, POD activity of high concentration up-regulated, decreased while the low concentration, MDA was opposite. On the long period of NaHCO3 salt stress, Nongda No.3 and Nongda No.4 of Cerasus humilises both showed highly tolerant against salt-stress.The plants from two provenaces, Lamagoumen and Huairou-tanghekou of Cerasus humilises in two year old, were selected as the materials, the natural drought stress in 50 days was used and several physiological characteristics was measured in order to analyze their physiological response to the stress. The results showed that the chlorophyll, carotenoid and MDA content, POD as well as SOD activity of 2 resources all changed during the stress. There were no significant difference between the stress treatment and the control among most of them except for chlorophyll content for Huairou-tanghekou provenace and POD activity for Lamagoumen resource. The electrolyte exosmosis percentage and MDA content of Labagoumen resource increased to the peak in the 32 days, but the electrolyte exosmosis percentage of Huairou-tanghekou resource incresased gradually in the early time of the treatment, and then stably went down, whose MDA content rised to the peak in the 44 days. The cytochrome content of 2 resources, no matter what was chlorophyll or carotenoid, increased sharply in the early 14 days, and decreased to the origin level for Lamagoumen resource, meanwhile, the content for Huairou-tanghekou resource also increased rapidly at the early stage of the treatment, and then fluctuated in the higher level to the end. POD activity gradually increased, and reached hightest value in the end of the treatment for both resources, but the POD activity level of Huairou-tanghekou resource was greatly higher than that of Lamagoumen provenace. The highest value of SOD activity of 2 resources was about 0.8 U·mg-1min-1, but it appeared in the 32 days for Lamagoumen resource, and in the 14 days for Huairou-tanghekou resource, and then decreased until the end of the experiment. Therefore, Huairou-tanghekou resource seemed to have stronger drought resistance than Lamagoumen resource.Cerasus humilises in Huangshandian resource were used as materials, whose several physiological characteristics by NaHCO3 stress, drought stress as well as NaHCO3 salt/drought stress in 25 days was measured in order to analyze their physiological response to the stress. The results showed that there existed significant difference in chlorophyll a, MDA and proline content among different treatments; No clear physiological variation in electrolyte exosmosis percentage, POD activity and proline content between the salt treatment and the control could be seen during the measuration, but not for drought and salt/drought treatment. The correlation analysis indicated that the same positive correlation was existed between salt and drought treatments, such as electrolyte exosmosis percentage, MDA content and POD activity,and a significant correlation among electrolyte exosmosis percentage,POD activity and MDA content by NaHCO3 stress, which meant there possibly existed the same physiological mechanism for Cerasus humilises by both of NaHCO3 salt and drought strees.There are significant difference in height, crown size, leaf shape among different resources of Cerasus humilis(Bge)Sok. The correlation analysis showed positive correlation between height and leaf length. The leaf width and leafstalk length are also positive correlation. However, relationship of leaf length with plant height, main branch length and crown width showed negative correlation. The negative correlation between leaf length and main branch length was significant.Soluble sugar and protein are significant difference between different resources of the Cerasus humilis(Bge)Sok. In this study, soluble sugar content in Tangshan and Nongda 3 were high during different growth period. However, soluble sugar content in Nongda 4 and Huangshandian were low. The soluble protein in different resources of the Cerasus humilis(Bge)Sok were similar to soluble sugar content. The primary results confirmed Tangshan and Nongda 3 resources are excellent according soluble sugar and protein content index.The analytic result from eight resources of the Cerasus humilis(Bge)Sok showed that 61 polymorphism sites can be amplified from a total of 80 individuals by 11 primers, the proportion of the polymorphic loci was 78.21%, the average of each primer over 5.5 polymorphic sites. The genetic diversity of the eight kinds of Cerasus humilis(Bge)Sok was estimated by Shannon indice:Tangshan>Huairou-labagoumen> No.5 of Nongda> No.4 of Nongda> No.3 of Nongda>Huairou-tanghekou>Yanqingjianjiadian>Huangshandian. There existed certain genetic differentiation in Cerasus humilis(Bge)Sok. The genetic variation among the populations was 25.32% of the total, and the other 74.68% distributed among individuals within populations. This indicated that the variation of Cerasus humilis(Bge)Sok mainly came from within populations. On the basis of the cluster analysis of genetic distance matrix, the eight kinds of Cerasus humilis(Bge)Sok can be divided into two categories:the genetic similarity among the resources of Huairou-labagoumen, Yanqingjianjiadian, Huairou-tanghekou> No.3 of Nongda,No.4 of Nongda and No.5 of Nongda are obvious; while the variation between Tangshan and Huangshandian were enormous with the others.
