Inner Mongolia Cashmere goat is one of the most famous native goat breeds due to its thin cashmere, and cashmere yield, strong adaptability as well as many other characteristics. To maximize use the excellent propagation potential of bucks.This study was aimed to find out the best fit dilution recipes and freezing programs of the semen cryopreservation for the Inner Mongolian Cashmere goats, and also lay the foundation for the practical application of semen cryopreservation of Inner Mongolian Cashmere goats. The results suggested as following:It was well-known that there is still a big gap between the cryopreservation of goat spermatozoa and the cryopreservation of bovine spermatozoa.In order to improve the quality of frozen-thawed goats semen,two ways including freezing methods and freezing extenders were studied in this present study. The spermatozoa parameters(including spermatozoa motion),acrosome integrity,membrane integrity , and DNA integrity were evaluated by flow cytometer (FCM) system,Fluorescein isothiocyanate-labeled pisumsation agglutinin (FITC-PSA) staining,JC-1 staining,hypo-osmotic swelling test (HOST) and so on.In this present study,fivee xperiments were carried out.1.The ram spermatozoa optimum cryopreservation procedure was established evaluated by the targets such as frozen-thawed semen quality:(1) Dilution rate is1:3of semen:extender,(2) Sperm diluted with single dilution method, (3) equilibrated at 5℃for 3 h. After equilibration, the straws were frozen in liquid nitrogen vapor, 4 cm above the liquid nitrogen, for 10 min. The frozen straws were thawed individually at 55℃for 6 s in a water bath for microscopic evaluation.2. In the nine formulations based on the further optimization of the basis to determine the best dilution D group tris-based extender (Tris 3.07 g/100 ml,citric acid 1.64 g/100 ml, glucose 0.5 g/100 ml, fructose 0.66 g/100 ml, egg yolk 15% (v/v), glycerol l5% (v/v): pH 6.8).3. In during the procession of cryopreservation of gaot semen,in the tris-based extender,add 62.5 mM trehalose gaot sperm can improve the effects of freeze-thaw,compared with the control group,and sucrose,and dextran. Trehalose significantly improved(P<0.05) the motility increased to 44.34%.and cryoprotectants comparative study showed that the incorporation of 5% glycerol significantly improved(P<0.05)the parameters of sperm motility.compared with DMSO, ethylene glycol,and propylene glycol. L-glutamic at concentrations of 5 mM significantly improved(P<0.05) sperm motility copared with the control with extender,and wih hypotaurine and cysteamine.In dilute solution by adding L-glutamic freezing the sperm motility rate were 48%.4. Sperm DNA damage has been associated with poor semen quality . Statistically significant negative correlations have been found between sperm motility and parameters such as defective acrosomes, AO, and TUNEL. Although there was a statistical correlation between AO- and TUNEL-positive spermatozoa, the investigation of DNA integrity using the TUNEL assay gave better results than AO staining. poor-quality semen has a greater percentage of spermatozoa with DNA fragmentation than good-quality semen. In our study, there was apositive correlation between sperm with defective acrosomes and apoptotic (as determined through both AO and TUNEL) spermatozoa. TUNEL and AO assays revealed that the freezethawing procedures also caused deterioration of DNA integrity. For TUNEL and AO staining, chromatin injury was the highest in post-thaw semen and differed significantly from diluted, cooled to 5℃, and equilibrated spermatozoa (P < 0.05). All cryoprotectants used in this study triggered apoptosis to some degree (P > 0.05).5 The objective of this research project was to develop an effective and plausible method to harvest testes and retrieve epididymal sperm from postmortem Cashmere goat deer that could be successfully cryopreserved for subsequent use in artificially insemination of does. And effect of goat epididymal sperm,on the role of toxic with bulbourethral gland secretion (BUS) contains phospholipase A combined with the egg yolk.
