The Cloning,Sequncing And Expressing Of Polyhedin Gene Of LdMNPV

The cloning, sequncing and expressing of polyhedin gene of LdMiNPV were studied. Ld.MNPV was purified and the DNA of LdMNPV was extracted. The PCR of LdMNPV was carried out with the primer 1 (5?CC CAT ATG CAC AAC TTT TAC AAC TAC 3?andprimer2(5 CC AAG CTT AGT ACG CCG GTC CTTG 3?,throughwhich the LdMNPV polyhedrin gene was cloned. The LdMNPV polyhedrin gene was cloned to I梀ector. The sequncing of LdMNPV polyhedrin gene showed that there were 735 bp in the ORF of LdMNPV polyhedrin gene in which 2bp(T, 378;C, 507) were different from the corresponding 2bp(C, 378;T, 507) in the published sequence of LdMNPV polyhedrin gene (Ian R. L. Smith, 1988). But the two corresponding codons codes the same amino acids, they are arginine and isoleucine respectively. The LdMNPV polyhedrin gene was expressed in. the strain B L 21( D E 3 )of F coil with the vector of pT7? plasmid.
