Anthurium Anthurium rank second only to tropical orchid as the most popular tropical flower with the high ornamental value and economic value. However, the research on Anthurium mainly focus on the tissue culture, cultivation and plant disease prevent and control. So far, research on coloration of anthurium has been reported rarely. The anthocyanin biosynthetic genes well characterized in antirrhinum, petunia, maize, Arabidopsis, grape and some other model species. Nowdays, the anthocyanin biosynthetic pathway was one of the best known pathways in dicot plants. But research in monocot plants, especially those have unusual organs like anthurium, were seldom.In order to analyze the molecular mechanism of coloration, this study used the 'Albama' wild spathe as materials; full-length cDNAs of 6 relevant structural genes in anthocyanin synthsis way were cloned. Then, anthocyanin accumulation of different colors in same growth stage spathes was measured. Finally, Semi-quantitative RT-PCR was performed to analyze expression characteristics of 6 genes related to anthocyanin synthsis in different spathes.The main result was listed as follows:1. Difference in phenotype:The differences of color in varieties were obvious.2. The anthocyanin accumulation showed that anthocyanin content had positive correlation with spathe color.3. Anthurium RNA was reverse transcripted into cDNA and the full-length cDNAs of 6 genes (AnCHS,AnF3H, AnDFR,AnFGT,AnLAR,AnANR) were amplified. Analysis of these 6 genes nucleic acid and mino acid were performed with bioinformatics. Their genes were predicted as well.4. The result of semi-quantitative RT-PCR showed that expression level of AnCHS,AnF3H,AnDFR,AnUFGT,AnLAR,AnANR were obviously different. AnCHS,AnF3H, AnDFR, AnUFGT and AnLAR expressed higher in the red color system and high anthocyanin content varieties, but AnANR expressed lower in them. The variety which had no anthocyanin content also had no genes expression.
