Studies On The Effective Ingredient And Pharmaceutical Action Of Compounds Solanum Lyratum (Thunb.) Injection

At present, bacterial or virus disease for animals begin to show some new characters during its outbreaks and epidemicity. Meanwhile, some untoward reactions on animals resulting from illegitimate use, and chemical residues in tissues of food animals, both severely hold back the improvement of farming. To overcome these shortcomings, compound Solarium Lyratum Thunb injection was prepared and studied, the result was as follows:(1) Compound Solarium Lyratum Thunb injection was made up of five traditional Chinese medicines, of which Solarium Lyratum Thunb was sovereign, indigowoad Root, Rhizoma coptidis, Herbal Tavaxaci were auxiliary, and Atralylodes Macrocephala koidz was adjuvant. According to the theory of veterinary and pharmacology of traditional Chinese medicine and the character of single medicine, the preparation was acquired by way of hydroalcoholic extracting and preparation regulations. The preparation was proved to be a brown-red clear pellucid fluid whose pH value was 5 + 0.5, and 1 ml of the preparation corresponded with 1 g of crude drug.(2) The qualitative analysis for the preparation were carried out by means of preparation of effective ingredient and filter paper chromatography. The results suggested that the preparation contained alkaloids and alkaloids salts, polysaccharides and glycosides, organic acids, etc. A uv spectrum method was established and employed in the quantitative determination of the contents of berberine, total polysaccharides and chlorogenic acids. The results indicated that the contents of the berberine, total polysaccharides and chlorogenic acids was 1.44mg/ml, 37.7mg/ml and 33.7mg/ml, respectively. The analysis for the methods above showed that the results were accurate, the percent recovery was high, the variation coefficient accorded with the demands and the data were stable.(3) With little white mice as materials, acute toxic experiments were carried out through the karber way. With the results, the semilethal dose of the preparation was91.5g/kg bw and its 95% of reliability was 18.2-21.1g/kg bw .Therefore, the preparation was basically no toxicity based on the standard of toxic evaluation. The pyrogen and irritation test was taken with white rabbit as materials respectively, the result suggested that there were no pyrogen and stimulation in the preparation. And a further study showed that intramuscular injection was safe and reliable.(4) The tube double dilution tests were used to measure the MIC and MBC of compound Solarium Lyratum Thumb injection. The results indicated that the MIC value of the preparation against streptococcus, staphylococcus, pseudomonas aeruginosa, solmonella paratyhpi and Escherichia coli was 1.56mg/ml, 1.56mg/ml, 12.5mg/ml, 12.5mg/ml, 25mg/ml, respectively and the MBC value was 3.12mg/ml, 3.12mg/ml, 25mg/ml, 25mg/ml, 50mg/ml, respectively. In vitro inhibitory experiments were used to measure antibacterial effect of compound Solarium Lyratum (Thunb) injection. The results of the experiment of antibacterial activity against staphyloccus accureus and streptococcus showed that the preparation had the same activity as that of both nicotinate norfloxacin and lincomycin (P>0.05), higher activity than that of maquindox(P<0.05); In the experiment of antibacterial activity against Escherichia coli, salmonella paratyphi and pseudomonas aeruginosa, the preparation had lower antibacterial activity than that of nicotinate norfloxacin. The results for in vitro antivirus test indicated that 100mg/ml of the preparation possessed notable antivirus activity against Newcastle disease virus and canine distemper virus, 10mg/ml of the preparation possessed faintish activity, 1mg/ml of the preparation possessed no activity.(5) In order to study the pharmaceutical action of the preparation, the effect of different doses of the preparation on the total number of white cells, the percentage of lymphocyte, the contents of total protein of serum and globulin, the index of thymus and spleen.
