Gene Differential Expression Analysis Of Adipose Tissue Pigs Treated With Clenbuterol Using Gene Chip

Clenbuterol, known as one of beta -adrenergic agonists is harmful to human being health after helping to improve procine meat quality traits. In order to understand which genes can directly or indirectly increase skeletal muscle mass and decrease body fat in livestock treated by clenbuterol, we analyzed gene differential expression of adipose tissues and muscle tissues of pigs with/without clenbuterol. In this study, we first created a cDNA microarray representing approximately 3500 pig genes for functional genomic studies. The array elements were selected from 6494 cDNA clones identified in a large-scale expressed sequence tag (EST) project. These cDNA clones came from normalized and subtracted porcine adipose tissue cDNA libraries. Sequence similarity searches of the 3426 ESTs represented on the array using BLASTN identified 2790 (81.4%) as putative human orthologs, with the remainder consisting of " novel" genes or highly divergent orthologs. By using the functional genomics analysis technical platform-cDNA microarray established by ourself, our main results are as follows:(1) Analyzed transcriptional differences between adipose tissues from Xiang pig and Large White at the gene expression level. Seventy-two different genes of the total 3426 genes on the array were differentially expressed two-fold or greater between Xiang pig and Large White. Among Seventy-two genes differentially expressed, we found sixteen are involved in lipid metabolism function; two are genes associated with cell organism and defence; three are relative with gene expression and protein synthesis and six genes were monitored to be relative with cell signal transduction. Other genes differentially expressed 2-fold or greater are thirty-four ESTs with no annotation and eleven ESTs representing novel genes.(2) Profiled transcripts expressed by adipose tissue of Chinese Xiang pig treated with/without clenbuterol during three months old. We obtained seventy genes induced and repressed and differentially expressed two-fold or greater. Among Seventy genes differentially expressed, we found thirteen are involved in lipid metabolism function; two are genes associated with cell organism and defence; eight are relative with gene expression and protein synthesis; two genes were monitored to be relative with cell signal transduction and nine are involved in cell structure and motility. Other genes differentially expressed 2-fold or greater are twenty ESTs with no annotation and sixteen ESTs representing novel genes.(3) Profiled transcripts expressed by adipose tissue of Chinese Xiang pig treated with/without clenbuterol during four months old. We obtained seventy-seven genes induced and repressed and differentially expressed two-fold or greater. Among Seventy-seven genes differentially expressed, we found fifteen are involved in lipid metabolism function; eight are relative with gene expression and protein synthesis; one gene was monitored to be relative with cell signal transduction and ten are associated with cell structure and motility. Other genes differentially expressed 2-fold or greater arethirty-two ESTs with no annotation and eleven ESTs representing novel genes.(4) Profiled transcripts expressed by adipose tissue of Large White treated with/without clenbuterol. We obtained one hundred and seven genes induced and repressed and differentially expressed two-fold or greater. Among one hundred and seven genes differentially expressed, we found twenty-five are involved in lipid metabolism function; two are relative with gene expression and protein synthesis; four are genes associated with cell organism and defence; five genes were monitored to be relative with cell signal transduction and one is involved in cell structure and motility.
