This study was conducted to assess the levels of genetic diversity of 55 kinds of alfalfa (Medicago sativa) with different origin including the cultivar from the domestic and some line from South Australia, North America and Europe. Alfalfa adapted to diverse the adaptation of alfalfa on diverse were tested.The comparison evaluation of alfalfa performance and characteristics are introduced in this study. The trial plots including 55 cultivars were established in Gansu Agricultural University. The growth speed, regeneration capacity as well as field yield were been tested. Different materials gave different growth speed, those green up earlier, grow quickly can achieve 4 times of cutting all the year round. The natural height and stem height were measured when cutting, average height were also tested before plant cease to prow in winter. The results show that there are significant differences in growth height, cutting height and fresh yield among different alfalfa lines. For example, Super7 can achieve 132.8cm height. Shanbei, Zhaodong show weak regeneration capacity, gave lower height at second and third cutting. The plot fresh yield in 2002 is 35.75 Kg/5m2-8.87 Kg/5m2, the average is 23.20Kg/5m2(46.4 t/ha). Sitel achieved the highest yield 35.75 Kg/5m2 (71.5 t/ha) ,the lowest one is Yanggao, only 8.87 Kg/5m2. The continuous precipitation caused delay for the first cutting, which influenced the second cutting yield of some lines grew quickly. The hay yield for testing materials varies between 9.68 Kg/5m ~2.29Kg/5m2, the average is 6.18Kg/5m2(12.4 t/ha).Sitel gave the highest plot yield, Xinjiang Daye, Prime, Derby and WL323 followed, the lowest one is Yanggao, which only got 8.87 Kg/5m2. There is large difference in fall dormancy of alfalfa. The height is 49.0cm~3.6cm before winter. L287, L90, LI73, L33 and Super7 all have higher fall height and winteractivity,but Longdong,Longzhong,Hexi appears lower fall dormancy.Technique of DNA molecularly marker for alfalfa was setup.In this study the primers which can find dependably polymorphic band were screened. The DNA polymorphism of the 55 alfalfa cultivars was evaluated by molecular markers. DNA fingerprinting of 55 alfalfa cultivars were described.Analysis system of conducted Randomly Amplified Polymorphic DNA (RAPD), Simple Sequence Repeat (SSR) and Inter-Simple Sequence Repeat (ISSR) on genomic DNA of alfalfa (Medicago sativa) were established. RAPD primers that have polymorphic bands were selected from offered abundance to do PCR amplify. We obtained dependably polymorphic bands by PCR amplify with screened 36 RAPD primers from 55 domestic and exotic alfalfa cultivars We obtained 120 polymorphic bands by PCR amplify with screened 32 RAPD primers,8 SSR primers and 12 ISSR primers from 55 domestic and exotic alfalfa cultivars. The results of SSR-PCR on alfalfa showed that there is large difference on SSR polymorphism among the alfalfa varieties. Polymorphism of alfalfa can be detected by fingerprinting assay. The ISSR which show abundantly polymorphism in fingerprint of alfalfa was steadier than RAPD. The genetic diversity of the 55 alfalfa cultivars was evaluated by RAPD,SSR and ISSR fingerprinting. The results of fingerprinting assay indicated that polymorphism of alfalfa can be detected and the alfalfa cultivars can be distinguished by 3 or 6 primers screened. The molecular markers can be effectively applied in genetic diversity of alfalfa.55 alfalfa were divided into some types by RAPD,SSR and ISSR analysis. The result of cluster showed there is small distance and near relative relation existed in the alfalfa of Longdong. Hexi and Longzhong. The Zhunger and Humeng had the same results. Tianshui alfalfa keeps big distance and far relation with the other domestic alfalfa. The Gannong3 and Gannongl had the same results with the above, and it showed the distinctive variety characters during the field experiments.Polyacrylamide gel electrophoresis on Alfalfa (Medicago sativa) was performed.
