The wild soybean is the wild relatives of cultivated soybean, it can not only provide excellent traits of cultivated soybean in crossbreeding, but also can screen high-quality gene in terms of drought tolerance and salt tolerance gene cloning, so it has an important significance for the optimization of the cultivation of soybeans.PEG6000of the experimental use of wild soybean seedlings to drought stress, the SOD gene of wild soybean by RT-PCR and RACE methods, and then the real-time quantitative PCR to detect the expression leve. According to the superoxide dismutase (SOD) published homologous full length cDNA sequences to design degenerate primers and a partial sequence of the open reading frame was amplified by PCR, the last we uesd RACE to obtain full-length sequence of the SOD gene named GsSOD. GsSOD the largest open reading frame of747bp, which encoded249amino acid residues, molecular weight of77.9Da protein, forecasting electricity point5.15. Hydropathy analysis showed GsSOD contain continuous hydrophobic fragment. And cultivated soybean (Glycine max), asparagus bean (Vigna unguiculata), cotton bean (Phaseolus lunatus), Lotus corniculatus (Lotus japonicus), Arabidopsis (Arabidopsis thaliana), grape (Vitis vinifera) and other plants compare related sequences, their superoxide dismutase of high homology, the sequence similarity of99%,98%,95%,93%,89%,96%,90%. We establish phylogenetic tree according to wild soybean and seven kinds of plant SOD gene sequence similarity, finally, it is consistent with traditional taxonomic status.
