The study focused on the expression of multigenes associated with lipid metabolism and the potential interact relationship among these genes and meat quality traits. Using Oligo microarray, 20 genes expression data from 6 to 14 weeks of age was obtained, a Bayesian network was constructed and the effect of gene's expression on meat quality traits of Beijing-you chicken was analyzed by different methods.The main innovation and results are as follows:1. We designed and fabricated an Oligo microarray, which has 48 genes subarray in one reflective slide for multi-sample hybridization at one time and the expression data of multigene was obtained in these samples. The innovated Oligo microarray reduces the experimental error caused by different hybridization. A special designed full-sib Beijing-you chincken population was used to reduce the effects of gentic background among samples.2. The expression data of 20 genes associated with lipid metabolism at 5 time series in growth period was analyzed. The average expression of UCP is 11.61, the highest in all these genes. Expressions of other genes are lower than GAPDH. Between expression of GH and body weight, the significant negative correlation (-0.36; P<0.05) was detected at 6 weeks of age, negative correlation (-0.36; p<0.10) at 8 weeks of age, and thereafter change to positive correlation (0.47; P<0.05) at 10 weeks of age.The significant negative correlations between leg intramuscular fat (IMF) content and several genes are found at 10 weeks of age. The middle correlation (0.31) of IMF between breast and leg is detected; the result indicated that the mechanism of fat accumulation differ between two kinds of muscles.3. A Bayesian network of these genes is constructed, which has 19 nodes and 28 lines. Spot14 a and Spot 14 β are start nodes and H-FABP connected 7 genes is the hub in the network. Connected 5 genes, ACC may play an important role in the network.4. The result of conditional probability and ANOVA inditcate that different expression of majority genes did not cause the difference of traits and significant result might not be identified if the value of conditional probability less than 50%. At 10 weeks of age, the IMF of the high expression groups of H-FABP and GH are lower than that of the control (P<0.05). The result of conditional probability is consistent with the result of ANOVA, but different from regression analysis.The results obtained from different methods indicate that the expression of many genes affect the some meat quality traits at 10 weeks of age. Among them, AMPK, LPL, H-FABP, GH and GHR have greater effect on meat quality. GH and GHR have negative effect on body weight and the IMF content of Beijing-you chicken was higher (P<0.01) in groups of high expression of LPL and GHR than the control.The effect of multigenes' expression on meat quality and the relationships among these genes were analyzed. Through this research, we reached the aim of the study that is to seek several analysis method for investigating the interact relationships among multigenes and traits. |
