Wheat germplasms, Octoploid Trititrigia Shannong TE263 and alien disomic addition line Shannong line 15, with resistance to powdery mildew , were developed from the progenies of common wheat Yannong15 ×Elytrigia intermedium (2n=6x=42, E1E1E2E2StSt), and were characterized by using of morphological, cyto-molecular genetics, biochemistry and molecular biological methods. And chromosome location of the resistance gene to powdery mildew from Elytrigia intermedium in these germplasms and its molecular markers were studied .The main results were as followes:1. The reports on the karyotypes of Elytrigia elongatum , Pseudowroegneria strigosa and Elytrigia intermedium were as follows. Both Elytrigia elongatum (2n=2x= 14= 10m+4sm(2SAT)) and Pseudowroegneria strigosa (2n=2x=14=8m+6sm (2SAT) ) belonged to 2 A type, while Elytrigia intermedium (2n=6x=42=16m+20sm(4SAT)+6st) belonged to 3B type. The results showed that E genome and St genome of Elytrigia intermedium had been evolved at large degree, compared with E genome of Elytrigia elongatum and St genome of Pseudowroegneria strigosa.2. Octoploid Trititrigia Shannong TE263(2n=28II=56) and alien disomic addition line Shannong line15(2n=22II=44), which were selected from the BC1F6 and BC1F5 of common wheat Yannong15×Elytrigia intermedium respectively, were identified by morphological and cytogenetics. The results of artificial inoculation of powdery mildew in Shannong TE263 and Shannong line 15 showed that they were resistant to powdery mildew and their resistant gene came from Elytrigia intermedium. Using 30 identification hosts and 21 differential isolates of E.graminis f. sp. Tritici to test the response type of resistance to powdery mildew revealed that the resistant response of gene in Shannong TE263 and Shannong line 15 was different from the resistance genes known. It indicated the resistant gene from Elytrigia intermedium is a probably new resistant gene to powdery mildew.3. Genomic in situ hybridization (GISH) using Elytrigia intermedium genomic DNA as probes and Chinese Spring DNA as block showed that Octoploid Trititrigia Shannong TE263 has 14 chromosomes of Elytrigia intermedium.
