SD rat and castrated male goat were used to study the effects of three kinds of isoflavones(7-hydroxyisoflavone and ipriflavone and daidzein) on the growth performance. The mechanism of soybean isoflavone improved the growth of male animal was studied. Soybean isoflavone could activite the cAMP/PKA signaling pathway and inhibited the activity of aromatase thoes lead to increasing the testosterone level and improving the growth of male animals. At the same time, we also probed into the absorption of soybean isoflavone in vitro and in vivo.1. Effect of three isoflavone phytoestrogens on the growth performance and mechanisms involvedTo study the effects of 7-hdroxyisoflavone and ipriflavone and daidzein on the growth performance in male and female rats by two series experiment and the mechanisms involved in the growth of male rats.1.1 Series I Effects of 7-hydroxyisoflavone and ipriflavone on the growth performance related endocrine function in ratsThe experiment was conducted with 30 rats of 28 days old to investigate the effect of 7-hydroxyisoflavone and ipriflavone at the dosage of 5 mg kg~-1 in diet for 6 weeks. Results showed that 7-hydroisoflavone and ipriflavone could increased the average daily body weight gain and decreased the feed intake/daily body gain, improved the ratio of RNA/DNA and the activity of glutamic Pyruvic transaminase and alkaline phosphatase and the level of testoterone . Compared with those male rat control, 7-hydroxyisoflavone and ipriflavone could increased the average daily body weight gain by 27.96 % (p<0.01) and 38.82%(p<0.01), feed intake/daily body gain was lowed by 27.71% and 38.34%, the level of testosterone increased by 252.34% (p<0.05) and 768.71% (p<0.01), respectively.7-hydroisoflavone and ipriflavone could increase the average daily body weight gain and decreased the feed intake/daily body gain. Compared with those female rat control,7-hydroxyisoflavone and ipriflavone could increase the average daily body weight gain by 86.38 % (p<0.01) and 17.12% (p<0.05), feed intake/daily body gain was lowed by 46.27% and 3.10%, respectively. 7-hydroisoflavone and ipriflavone had no significant effect on the level of estradiol and the content of urea-N and the activity of Pyruvic transaminase. Those results indicate that 7-hydroxyisoflavone and ipriflavone could improve the growth both male and female in rat and this function could have relation to the protein metabolism.1.2 Series II Effects of 7-hydroxyisoflavone and ipriflavone and daidzein on the growth performance and mechanisms involved in ratEighty growing rat were allotted randomly into four groups and in half male and female, feed diets supplement with 7-hydroxyisoflavone and ipriflavone and daidzein of 5 mg kg~-1 in experiment and 0 mgkg~-1 in control group for 6 weeks. Result showed that 7-hydroxyisoflavone and ipriflavone and daidzein could increase the average daily body weight gain and decreased the feed intake/daily body gain of male rat. 7-hydroxyisoflavone and daidzein could decreased the content of urea-N (p<0.01) and improved the content of growth hormone (p<0.01) in serum of male rats, meanwhile ipriflavone had no effects on the content of urea-N and growth hormone. At the same time, 7-hydroxyisoflavone and ipriflavone and daidzein could increase the content of testosterone and inhibited the activity of aromatase in testis of male rat. Compared with those male rat control , 7-hydroxyisoflavone and ipriflavone and daidzein were increased the content of testosterone by 73.87% (p<0.01) and 371.52 (p<0.01) and 112.67 (p<0.01), the activity of aromatase was inhibited by 66.47%(p<0.01 )and 57.64%(p<0.01 )and 12.32%(p<0.01), respectively.7-hydroxyisoflvone and ipriflavone could increase the average daily body weight gain and decreased the feed intake/daily body gain of female rat. Daidzein could decrease the average daily body weight gain by 4.13% (p>0.05) compared with female rat control.
