Clostera anachoreta is a major leaf worm of aspen and may be a good candidate for biological control of C. anachoreta. In this paper we studies ClanGV basic characterization, identified the gene map of granulin N-terminus region, did some research on Helicoverap argimra Peritrophic Membrane (PM). The detailed content as follows: Chapter 1 of this thesis was a general introduction of baculoviruses ,granulovirses and insect PM research. In chapter 2, ClanGV was analyzed for multiplication,virus purification,genome extraction,morphology,restriction endonuclease digestion and ascertain baculoviral enhancin homologue. It was fundamental research about ClanGV for further functional genome research. In chapter 3 the granulin N-terminal 10 kb area was identified and the included genes were analyzed using biosofts. The gene order in sequenced fragments was compared with other GVs. the results indicated ClanGV and ClGV, CpGV, PoGV, AoGV have the similar gene order and maybe more related. In chapter 4, the ClanGV granulin gene was cloned,sequenced and analyzed. The granulin protein was extracted and the rabbit was immunized to get the antibody. Western blot analysis indicated ClanGV granulin was more related to CaLGV and HaNPV. Chapter 5 reported the morphology, structure and component of Helicoverpa armigera normal PM, and the effect of Congo Red (CR) on the growth was also studied. The results indicated: the PM of H. armigera resembled typeâ… . The insect couldn't synthesize intact PM after consumed the artificial diet contained 2% CR, and the breakage could be resumed by the diet without CR. 1%CR could enhance the sensitivity of larva to HaNPV.
